Experimental Study on the Protective of Sangqi Shouwu on Cerebral Ischemia-Reperfusion Injury in Rats

目的:探究桑芪首乌方(Sangqi Shouwu,SQSW)对大鼠缺血再灌注损伤大脑的保护作用.方法:构建大脑中动脉闭塞(MCAO)大鼠模型,将大鼠分成5组,分别为:假手术组、模型组、SQSW低剂量组(1.2g生药/mL)、SQSW中剂量组(2.4g生药/mL)和SQSW高剂量组(4.8g生药/mL).根据Zea Longa法进行神经功能评分,采用干湿称重法检测大鼠脑含水量变化.采用TTC染色法检测大鼠脑梗死体积,采用HE染色法检测大鼠海马组织神经元病理学改变,采用TUNEL染色法检测大鼠脑组织细胞凋亡,采用Western blot法检测大鼠脑组织中相关蛋白的表达.结果:与模型组大鼠比较,SQSW干预后能够显著降低大鼠的神经功能评分,减少大鼠的脑含水量和大鼠脑梗死率.与模型组相比,SQSW加药组大鼠脑组织海马CA1区神经元病理学改变明显减少,脑组织细胞凋亡率显著降低,SQSW加药组大鼠患侧脑皮质组织中Notch1、Hes1、Bax、Caspase-3、Caspase-9蛋白相对表达量均显著降低.结论:SQSW对大鼠缺血再灌注损伤大脑具有一定的保护作用,其作用机制与抗凋亡及Notch信号通路相关.
Objective To investigate the protective effect of Sangqi Shouwu(SQSW)on cerebral ischemia-reperfusion injury in rats.Methods The middle cerebral artery occlusion/reperfusion(MCAO)rat model was constructed and divided into 5 groups:sham operation group,model group,low dose SQSW group(1.2g crude drug/mL),medium dose SQSW group(2.4g crude drug/mL)and high dose SQSW group(4.8g crude drug/mL).Zea Longa assay was used to score neu-ral function,and dry-wet weighing method was used to detect the changes of brain water content.The factors related to oxidative reaction in rats were detected by free immunoassay,the volume of cerebral infarction was detected by TTC staining,the pathological changes of hippocampal neurons were detected by HE,the apoptosis of brain tissues was detected by TUNEL,the expression of re-lated proteins in rat brain tissues was detected by Western blot.Results Compared with model group,SQSW intervention can significantly reduce the neurological function score of rats,reduce the brain water content of rats.Compared with model group,the neuronal pathological changes in hippo-campal CA1 area of rats in SQSW group were significantly reduced,and the apoptosis rate of brain cells was significantly reduced.The relative expression levels of Notch1,Hes1,Bax,Caspase-3 and Caspase-9 proteins in the affected cerebral cortex of SQSW group were significantly decreased.Conclusion SQSW has a protective effect on cerebral ischemia-reperfusion injury in rats,and its mechanism is related to anti-apoptosis and Notch signaling pathway.
