摘要

目的:研究天智颗粒对Aβ25-35诱导的大鼠嗜铬细胞瘤PC12细胞凋亡的作用及其机制.方法:用不同浓度天智颗粒或天智颗粒+LY294002干预Aβ25-35处理的PC12细胞,通过MTT检测细胞活力,流式细胞仪检测细胞凋亡和氧应激,酶标仪检测Caspase 3活性和丙二醛(MDA)含量,免疫印迹法检测Bcl2/Bax、p-AKT/AKT蛋白水平.结果:天智颗粒剂量依赖地改善Aβ25-35诱导的PC12细胞活力下降,抑制其凋亡,降低Cas-pase 3活性,上调Bcl2/Bax表达,减少氧应激和MDA的生成,同时促进PI3K/AKT信号的活化.而PI3K抑制剂LY294002干预显著地逆转天智颗粒的这些有益作用.结论:天智颗粒能有效地保护Aβ25-35诱导的PC12细胞凋亡,其机制可能与激活PI3K/AKT信号相关.

Abstract

Objective:To explore the effect and mechanism of Tianzhi granules on Aβ25-35-induced pheochromocytoma PC12 cells apoptosis.Method:PC12 cells were induced by Aβ25-35 in the pres-ence or absence of different dose of Tianzhi granules without or with LY294002.The cell viability was determined by MTT,apoptosis and oxidative stress were measured by flow cytometry,caspase 3 activity and MDA content were detected by microplate reader.The levels of Bcl2,Bax,p-AKT and AKT protein were determined by western blotting.Results:Tianzhi granules dose-dependently im-proved Aβ25-35-induced PC12 cells apoptosis,as indicated by increased cell viability,decreased cas-pase 3 activity,as well as upregulating Bcl2/Bax.Furthermore,Tianzhi granules dose-dependently in-hibited Aβ25-35-induced oxidative stress and MDA content but activated PI3K/AKT signal pathway,and the inhibitor of PI3K/AKT,LY294002,reversed the beneficial effects of Tianzhi granules in Aβ25-35-primed PC12 cells.Conclusion:Tianzhi granules could effectively protect from Aβ25-35-primed PC12 cells,which might be mediated by activating PI3K/AKT pathway.
