益母草水苏碱联合交联透明质酸钠调节兔宫腔粘连模型纤维化的研究

目的:评估益母草水苏碱联合交联透明质酸钠治疗兔宫腔粘连的效果并探讨其机制.方法:将新西兰兔随机分为假手术组、模型组、实验组三组,采用机械损伤法制作宫腔粘连模型.实验组术中予益母草水苏碱联合交联透明质酸钠灌注宫腔,术后益母草水苏碱腹腔注射治疗.2周后观察子宫组织形态学变化、比较子宫内膜腺体计数、血管计数及纤维化面积,蛋白印迹法检测Smad4、转化生长因子-β1(TGF-β 1)、基质金属蛋白酶组织抑制因子(TIMP)-1、基质金属蛋白酶(MMP)-9及血管内皮生长因子(VEGF)1蛋白含量.结果:与假手术组相比,模型组子宫腺体计数与血管计数明显减少(P<0.05),子宫内膜纤维化面积比显著增加(P<0.05),子宫组织中Smad4、TGF-β1及TIMP-1蛋白表达明显上升(P<0.05),MMP-9蛋白水平显著下降(P<0.05).与模型组相比,实验组子宫内膜的腺体及血管的数量增加,纤维化面积减少(P<0.05),实验组子宫Smad4、TGF-β1、TIMP-1的蛋白含量明显下降(P<0.05),MMP-9及VEGF1蛋白水平明显升高(P<0.05).结论:益母草水苏碱联合交联透明质酸钠具有改善宫腔粘连的作用,机制可能与抗纤维化及血管新生有关.
Study of Stachyrine Hydrochlorid Combined with Cross-linked Sodium Hyaluronate Gel in the Fibrosis of Intrauterine Adhesions Rabbit Model
Objective:Evaluate the therapeutic effect of stachyrine hydrochlorid combined with cross-linked so-dium hyaluronate gel on intrauterine adhesions(IUA)in rabbits and the relevant mechanism of action.Meth-ods:Rabbits were randomLy divided into three groups:sham-operated group、model group、experimental group.IUA rabbit model was induced by mechanical damages.Experimental group were perfusion stachyrine hy-drochlorid(Sta)combined with cross-linked sodium hyaluronate gel to the uterine cavity during the surgery,and injected Sta after the surgery.The histological changes of the uterus were observed after 2-week medication,compared with the number of the endometrial glands and blood vessel mass,the fibrosis area in each group.The expressions of Smad4、TGF-β1、TIMP-1、MMP-9 and VEGF1were detected using West blot.Resuits:Compared with sham-operated group,the number of the endometrial glands and blood vessel mass were reduced.the fibrosis area were increased(P<0.05),the expression of Smad4、TGF-β1 and TIMP-1 were elevaled the expression of MMP-9 decreased in model group(P<0.05).Compared with model group,he num-ber of the endometrial glands and blood vessel mass were increased.the fibrosis area were reduced(P<0.05),the expression of Smad4、TGF-β1、TIMP-1and VEGF1 were decreased,the expression of MMP-9 elevaled in experimental group(P<0.05).Conclusion:Sta combined with cross-linked sodium hyaluronate gel can improve endometrial fibrosis in IUA rabbits,and its mechanism may be related to the anti-fibrotic ac-tions and neovascularization.
