内生真菌群落在不同植物的不同组织中具有不同的组成和分布,采用ITS高通量测序技术比较4种不同桉树叶片、果实和枝条内生真菌群落组成和多样性差异.4种桉树(Eucalyptus spp.)内生真菌群落结构分析结果表明,子囊菌门(Ascomycota)和担子菌门(Basidiomycota)是所有样本的优势菌门,但在不同样本中的相对丰度有差异.生物多样性分析结果显示,在同一组织和不同组织中托里桉和柠檬桉的内生真菌群落均相似,而窿缘桉与无性系广9在同一组织中内生真菌群落相似,在不同组织中内生真菌群落差异较大.4种桉树在枝条中的多样性指数无明显差异,而窿缘桉果实、无性系广9的叶片和果实内生真菌群落的Chao1、Observed species和PD whole tree指数均显著高于托里桉和柠檬桉.LEfSe分析表明,柠檬桉(枝条和果实)和窿缘桉(叶片和果实)中的差异物种分布在子囊菌门,托里桉(果实)和无性系广9(叶片和枝条)中的差异物种在子囊菌门和担子菌门中均有分布.桉树中蕴含了丰富的内生真菌菌群,因树种和组织的不同而存在较大差异,无性系广9叶片和窿缘桉果实部位拥有更丰富的内生真菌群落,本研究为后续桉树内生真菌资源的开发和利用提供重要依据.
Community composition and diversity of endophytic fungi in leaves,fruits and branches of different Eucalyptus spp.
Endophytic fungal communities have different compositions and distributions in different tis-sues of different plants.The community composition and diversity of endophytic fungi in leaves,fruits and branches of four Eucalyptus species were compared by ITS high-throughput sequencing technique.The results of endophytic fungal communities structure analysis of four Eucalyptus species showed that As-comycota and Basidiomycota were the dominant phyla in all samples,but their relative abundance varied in different samples.The results of biodiversity analysis illustrated that the endophytic fungal communities of Eucalyptus torelliana and E.citriodora were similar in the same tissue and in different tissues,while E.exserta and G9 had similar endophytic fungal communities in the same tissue but with differences in different tissues.The diversity indices of the four Eucalyptus species in branches were not significantly different,while the Chaol,Observed species and PD whole tree indices of endophytic fungal communities were significantly higher in the fruits of E.exserta and G9 than in E.torelliana and E.citriodora.LEfSe analysis showed that the differential species in E.citriodora(branches and fruits)and E.exserta(leaves and fruits)were distributed in the Ascomycota,while those in E.torelliana(fruits)and G9(leaves and branches)were distributed in both Ascomycota and Basidiomycota.Eucalyptus spp.contains abundant endophytic fungal flora,which varies greatly in different species and tissues.There are more abundant endophytic fungi community in the leaves of G9 and fruits of E.exserta.This study provides an important basis for the development and utilization of endophytic fungal resources in Eucalyptus spp..
