Expression of NLRP3 and Its Clinical Significance in Gastric Cancer
[Objective]To investigate the expression of NLRP3 and its correlation with prognosis in gastric cancer.[Methods]The data of NLRP3 mRNA expression in 567 gastric cancer patients were obtained from GEO database.A total of 112 cancer tissue and 82 pericancerous normal tis-sue samples were collected from gastric cancer patients who received surgical treatment in Kun-shan Hospital from January 2014 to August 2017,and the expression level of NLRP3 protein was detected by immunohistochemistry.The correlation between NLRP3 protein and mRNA expression levels and patient prognosis was analyzed by univariate and multivariate Cox analysis.[Results]Among the 567 gastric cancer patients from the GEO database,104 cases(18.34%)had low ex-pression of NLRP3 mRNA and 463 cases(81.66%)had high expression.Univariate Cox regression analysis showed that TNM stage,age,lympho-vascular invasion,and NLRP3 mRNA level were correlated with the prognosis of gastric cancer patients(all P<0.05).Multivariate Cox regression analysis showed that low expression of NLRP3 mRNA was an independent risk factor for prognosis(HR=1.39,95%CI:1.04~1.87,P=0.028).Among 112 clinical gastric cancer patients,the mean immunohistochemical score of NLPR3 in cancer tissues was lower than that in pericancerous tis-sues(3.49±3.16 vs 5.54±3.51,P<0.001).There were 18 cases with negative expression of NLPR3 protein and 94 cases with positive expression.The expression of NLRP3 protein was correlated with age,T stage,and TNM stage(all P<0.05).The prognosis of patients with negative NLRP3 protein expression was significantly worse than that of positive expression patients(HR=2.34,95%CI:1.04~5.30,P=0.041).[Conclusion]Low expression of NLRP3 is a risk factor for the poor prognosis of gastric cancer patients,which may be used as predictive marker for prognosis of gastric cancer.
