Circ_0001946靶向miR-671-5p调控Wnt/β-catenin通路对结直肠癌细胞恶性进展的影响

[目的]探讨circ_0001946对结直肠癌细胞恶性进展的影响.[方法]培养结直肠癌HCT116、SW480、Colo320、HT29 细胞和正常结直肠 CCC-HIE-2 细胞,实时定量 PCR 检测 circ_0001946、miR-671-5p表达量.miRcode和双荧光素酶报告实验分析circ_0001946和miR-671-5p的关系.circ_0001946、miR-671-5p在HCT116细胞中的表达量被上调或下调后,分别检测HCT116细胞生长活性、细胞凋亡率及细胞迁移能力.免疫蛋白印迹法检测Bcl-2、Bax、Caspase-3和Caspase-9蛋白表达及Wnt/β-catenin信号通路蛋白表达.[结果]HCT116(4.76±0.47)、HT29(2.85±0.57)、SW480(2.75±0.62)、Colo320(3.12±0.68)细胞中circ_0001946 表达水平高于 CCC-HIE-2 细胞(1.24±0.09)(t=7.238、8.129、6.893、6.875,P均<0.05).下调circ_0001946表达显著降低了 HCT116细胞增殖(P<0.05)和迁移能力(P<0.05),促进了细胞凋亡(P<0.01);circ_0001946靶向miR-671-5p;circ_0001946表达被上调或miR-671-5p表达被下调后,HCT116细胞增殖和迁移能力显著上升,细胞凋亡率明显降低,Wnt/β-catenin信号通路被激活.[结论]circ_0001946靶向miR-671-5p激活了Wnt/β-catenin信号通路,促进了结直肠癌细胞恶性进展.
Circ_0001946 Promotes Malignant Progression of Colorectal Cancer Cells by Activating Wnt/β-catenin Pathway Through miR-671-5p
[Objective]To investigate the effect of circ_0001946 on the malignant progression of colorectal can-cer cells and its mechanism.[Methods]The expression levels of circ_0001946 and miR-671-5p were detected in human colorectal carcinoma HCT116,SW480,Colo320,HT29 cells and normal colorectal CCC-HIE-2 cells by RT-qPCR.The relationship between circ_0001946 and miR-671-5p was analyzed by miRcode and du-al luciferase reporting assay.After circ_0001946 and miR-671-5p expression in HCT116 cells were up-regulat-ed or down-regulated,the growth activity,apoptosis rate and migration ability of HCT116 cells were detected respectively.The expression of Bcl-2,Bax,Caspase-3 and Caspase-9 and the proteins of Wnt/β-catenin sig-naling pathway were detected by Western blot.[Results]The expression levels of circ_0001946 in HCT116(4.76±0.47),HT29(2.85±0.57),SW480(2.75±0.62),and Colo320(3.12±0.68)cells were significantly higher than those in CCC-HIE-2 cells(1.24±0.09)(t=7.238,8.129,6.893,6.875,all P<0.05).Down-regulation of circ_0001946 significantly reduced the proliferation(P<0.05)and migration(P<0.05)of HCT116 cells,and promoted cell apoptosis(P<0.01).The dual luciferase reporting assay showed that circ_0001946 targeted miR-671-5p.Up-regulation of circ_0001946 or down-regulation of miR-671-5p significantly enhanced proliferation and migration of HCT116 cells,decreased apoptosis rate,and activated Wnt/β-catenin signaling pathway.[Conclusion]The study indicates that circ_0001946 may promote malignant progression of colorectal cancer cells by activating the Wnt/β-catenin signaling pathway through miR-671-5p.
