Effects of Dandelion Extract on Proliferation and Biological Activi-ties of Gastric Cancer Cells and Its Mechanism
[Objective]To investigate the effects of dandelion extract(DE)on proliferation and biological activities of gastric cancer(GC)cells and its molecular mechanism.[Methods]DE was prepared by dis-tillation and freeze-drying techniques,and gastric cancer BGC-823 and AGS cells were treated with dif-ferent concentrations(0.280,0.560,1.125 μmol/L)of DE.The proliferation,apoptosis,cell cycle,mi-gration,and invasion of gastric cancer cells by MTS method,Transwell assay,and flow cytometry.The mRNA and protein expression of proliferating cell nuclear antigen(PCNA),B-cell lymphoma extra-large(Bcl-xl),B-cell lymphoma-2(Bcl-2),matrix metalloproteinase 9(MMP-9),E-cadherin,IL-6,IL-8 and IL-1β were detected by RT-qPCR and Western blotting,respectively.[Results]Compared with the control group,the proliferation of the DE treated groups decreased in a time and concentration-depen-dent manner(all P<0.05),and the apoptosis rate increased significantly with the increase of DE concen-tration(P<0.05).The proportion of G0/G1 phase cells was higher and G2/M phase cells was lower in the DE treated groups(all P<0.05).Compared with the control group,the migration and invasion abilities of cells in the DE treated groups decreased with increasing DE concentration(P<0.05),and the protein and mRNA expression of PCNA,Bcl-xl,Bcl-2,MMP9,E-cadherin,IL-6,IL-8 and IL-1 β were sig-nificantly reduced(all P<0.05).[Conclusion]DE can inhibit the proliferation,migration and invasion of gastric cancer cells,promote cell apoptosis and induce cell cycle arrest,which may be related to its regulation of the expression of related genes and proteins.
