Determination of Protocatechuic Aldehyde and Protocatechuic Acid in the Different Medicinal Parts of Thladiantha dubia Bunge
[Objective]To establish an assay method to determine the content of protocatechuic aldehyde and protocatechuic acid in the roots and fruits of Thladiantha dubia.[Method]Protocatechuic aldehyde and protocatechuic acid were separated and analyzed by Discovery C18 column(4.6 mm×250 mm,5 μm),mobile phase was methanol-5%phosphoric acid(0-40 min,methanol:20%～100%)with gradient elution at the flow rate of 1 mL/min and the detection wavelength was set at 280 nm.[Result]Protocatechuic aldehyde had good linear relationship between 0.003-0.052 mg/mL(r=1.000 0),the average recovery was 100.6%and RSD was 1.27%;protocatechuic acid had good linear relationship between 0.002-0.042 mg/mL(r=1.000 0),the average recovery was 100.6%and RSD was 1.76%.[Conclusion]The method is simple,sensitive and reliable to carry out and control the quality of protocatechuic aldehyde and protocatechuic acid in the roots and fruits of T.dubia.
