大葱离体雌核培养及植株再生的研究

[目的]研究大葱胚状体及其再生植株的诱导方法.[方法]以大葱花蕾为试材,利用正交试验法对大葱离体雌核培养进行了相关研究.[结果]6-BA、NAA和2,4-D对诱导雌核出胚有极显著影响.[结论]添加 6-BA 2.5 mg/L、NAA 0.04 mg/L、2,4-D 1.0 mg/L为最优诱导培养基,雌核培养再生植株成活率可达 78.2%.
Study on Vitro Female Culture and Plant Regeneration in Allium fistulosum L.
[Objective]To study the induction of embryoid and regenerated plants from buds of Allium fistulosum L..[Method]In vitro gynogenetic culture of A.fistulosum L.was studied by orthogonal test.[Result]The results showed that 6-BA,NAA and 2,4-D had significant effects on gynogenesis.[Conclusion]The optimum induction medium was 6-BA 2.5 mg/L,NAA 0.04 mg/L and 2,4-D 1.0 mg/L.The survival rate of regenerated plants by gynogenetic culture can reach 78.2%.
