Isolation and promoting characteristics of the rhizospheric bacteria of Camellia oleifera Abel
[Objective]In order to lay the foundation for the development of specialized microbial fertilizer for Camellia oleifera Abel and improve the quality and yield of Camellia oil.[Method]Taking the rhizosphere soil of C.oleifera as test material,using PVK plate detection method,Alexander plate detection method,CAS plate detection method,JNFB culture medium detection method,and Salkowski colorimetric method to respectively to test phosphorus solubilization ability,potassium solubilization ability secreting,secreting iron ability,nitrogen fixation ability and producing IAA ability of 20 bacteria randomly selected from the rhizosphere soil of C.oleifera.The phosphorus solubilization ability and potassium solubilization ability of bacterium YC10 from C.oleifera rhizosphere were quantitatively determined using Mo-Sb Anti spectrophotometric method and flame atomic absorption spectrometry respectively.Molecular identification of strain YC10 was carried out using 16S rDNA.[Result]The results showed that among the 20 selected bacteria,6(30%)had the ability to dissolve phosphorus,6(30%)had the ability to dissolve potassium,10(50%)could produce ferritin,7(35%)had the ability to fix nitrogen,and up to 18(90%)had the ability to produce IAA.There were 14 strains with 2 or more growth promoting properties,accounting for 70%of the total isolated bacterial strains.Among them,strain YC10 had multiple growth promoting properties such as phosphorus solubilization,potassium solubilization,iron carrier production,IAA production,and nitrogen fixation.The effective phosphorus content in the fermentation broth of strain YC10 reaches its maximum of 68.09 μg·mL-1,with a phosphorus solution rate of 5.44%at 60 hours,its fermentation liquid soluble potassium content was 4.43 μg·mL-1,with a potassium solution rate of 31.63%at 72 hours.Strain YC10 was belonged to Burkholderia sp.by 16S rDNA.[Conclusion]There are many bacteria with growth promoting properties in the rhizosphere of C.oleifera,among which strain YC10 has multiple growth promoting properties such as phosphorus and potassium solubilization,iron carrier production,IAA production,nitrogen fixation,etc.It has the potential to develop specialized microbial fertilizer for C.oleifera.
