Phenotypic traits and ISSR genetic diversity analysis of poplar clones
[Objective]The phenotypic traits and molecular marker diversity were analyzed to explore the genetic diversity of 62 poplar clones and to lay a foundation for further genetic improvement of poplar.[Method]Genetic diversity of poplar clones was analyzed using 12 phenotypic traits(ground diameter,seedling height,leaf area,leaf length,leaf width,petiole length,chlorophyll,number of lateral branches,leaf thickness,total leaves per plant,leaf dry mass,leaf moisture content)and ISSR markers.Diversity index was calculated using PopGen 32,variance analysis among phenotypic traits was performed by SPSS 16.0 and UPGMA cluster analysis was performed for each clone by NTSYS2.10E software.[Result]The coefficient of variation of the number of lateral branches,the number of leaves per plant,leaf dry mass,leaf area and ground diameter reached more than 10%,and the coefficient of variation of petiole length,leaf length and seedling height reached more than 8%.Using 5 ISSR primers to detect 62 percentage of poplar polymorphic band was 89.04%on average,genetic diversity degree with an average of 0.407 4,Shannon diversity index(I)an average of 0.530 4.The morphological and molecular marker cluster maps constructed by UPGMA method showed a tendency of clustering into one class,but there were also great differences.The clustering analysis of phenotypic traits was mainly based on the higher similarity of leaf related traits,which were clustered into a group.Molecular marker clustering mainly showed the rule that the closer the clonal relationship was,the easier it was to cluster into a group.[Conclusion]The phenotypic traits of poplar clones were highly differentiated and had rich genetic diversity.The results provided a scientific basis for the improvement,germplasm innovation and diversified development and utilization of poplar germplasm resources.
