13个紫薇品种花粉形态及花粉活力分析

[目的]研究紫薇品种间的花粉形态特征及花粉活力差异,为后续紫薇品种杂交育种工作提供理论参考。[方法]通过扫描电子显微镜对13 个紫薇品种的雄蕊群花粉粒表面形态特征进行观察研究,每个品种随机选取 10 粒花粉进行统计分析,同时利用固体培养基对 13 个紫薇品种的花粉活力进行测定。[结果]13 个紫薇品种长短雄蕊花粉粒均呈长球形,有3条萌发沟,裂至接近两极,萌发沟间未连接,在极面观花粉粒呈钝三角形,均属于N3P4C5 型花粉。紫薇品种间花粉粒外壁纹饰、极轴长、赤道轴长、萌发沟特征及花粉粒体积大小均差异显著,其中花粉大小为 538。91~1 402。40 μm2,极轴长为 30。51~46。31 μm,赤道轴长为 17。35~31。49 μm,萌发沟长为 18。38~34。36 μm,萌发沟脊宽为 5。72~11。34 μm。花粉外壁主要包括孔穴状、脑皱状及皱波状 3 种纹饰,13个试验品种长雄蕊花粉粒相比于短雄蕊花粉粒,其花粉粒表面更为光滑饱满。花粉活力测定结果以'紫精灵'紫薇花粉活力最强,花粉活力为93。94%,其中'湘韵'紫薇为不育品种,其不育类型表现为雄性不育,花粉不萌发。[结论]紫薇不同品种的花粉粒形态特征和花粉粒表面纹饰可以作为紫薇品种分类鉴定的参考指标之一,本试验中的13 个紫薇品种除不育品种'湘韵'紫薇外,其他试验品种表现出较高的花粉活力,均可作为紫薇品种杂交育种工作材料。
Analysis of pollen morphology and pollen viability of thirteen Lagerstroemia indica varieties
[Objective]The study of pollen morphological characteristics and pollen vigor differences among varieties provides a good theoretical reference for the subsequent cross-breeding work of Lagerstroemia indica varieties.[Method]The surface morphology of the pollen grains of the stamen cluster of 13 L.indica varieties was observed and studied by scanning electron microscope,and 10 pollen grains of each varieties were randomly selected and statistically analyzed,while the pollen viability of the 13 L.indica varieties was determined by using solid culture medium.[Result]The pollen grains of the 13 L.indica varieties with short and long stamens were oblong spherical in equatorial view,with three germination grooves,cleft to close to the poles,not connected between the germination grooves,and obtusely triangular in outline in the polar view,and all of them belonged to the N3P4C5 type of pollen.The outer wall ornamentation of pollen grains,polar axis length,equatorial axis length,germination furrow characteristics and pollen grain volume size varied significantly among L.indica,with pollen size ranging from 538.91 to 1 402.40 μm2,polar axis length from 30.51 to 46.31 μm,equatorial axis length from 17.35 to 31.49 μm,germination furrow length from 18.38 to 34.36 μm,and the width of the germination furrow ridge from 5.72 to 11.34 μm.The outer wall of pollen mainly included three kinds of decorations:pore-like,brain-wrinkle-like and wrinkle-wave-like,and the surface of pollen grains of 13 test varieties with long stamens was smoother and fuller than that of pollen grains with short stamens.The results of pollen vigor determination showed that L.indica'Zijingling'had the strongest pollen vigor,with a pollen germination rate of 93.94%,among which L.indica'Xiangyun'was a sterile variety,whose sterility type was male sterility,and the pollen did not germinate.[Conclusion]Morphological characteristics of pollen grains and surface ornamentation of pollen grains of different varieties of L.indica can be used as one of the reference indexes for the classification and identification of L.indica varieties.The 13 varieties of L.indica in this experiment,except for the sterile variety of L.indica'Xiangyun',the other varieties of the experiment showed high pollen viability,and all of them can be used as the materials for the subsequent work of cross-breeding of L.indica varieties.
