Reconstruction of phylogenetic relationship of Asian Caryodaphnopsis(Lauraceae)based on nuclear genome
[Objective]Caryodaphnopsis Airy Shaw(Lauraceae)is rich in seed oil,and its fruit and wood have great development potential.Due to the small differences between species, the confusion of interspecific relationships and the small number of existing specimens, people have little understanding of the genus, which greatly hinders the development and utilization of the species of Caryodaphnopsis. Therefore,it is urgent to construct the phylogenetic relationship of the Asiatic group of Caryodaphnopsis.[Method]In order to further understand the Asian group of Caryodaphnopsis, the ribosomal genome(nrG)sequences of 44 Caryodaphnopsis plants were obtained by second-generation sequencing technology to construct a highly supported phylogenetic relationship, and comprehensive phylogenetic results and morphological traits comparative demonstration.[Result]Using nrDNA sequence matrix to construct ML tree and Bayesian tree, the grouping of ML tree and Bayesian tree was completely consistent.By comparing the ITS tree,it was found that the phylogenetic structure except for C.tonkinensis and C.latifolia was basically the same.C.laotica,C.metallica and C.hekouensis were still clustered together under a large branch, and C.latifolia and C.tonkinensis were also clustered together. The ITS sequence and nrDNA sequence tree showed that the genetic relationship between C.latifolia and C.tonkinensis was close. The genetic relationship of C.laotica,C.hekouensis and C.metallica was close, and it was speculated that hybridization or gene exchange occur between the two groups of species.Based on the comprehensive morphological traits,44 samples were divided into 9 groups. They were C.henryi,C.poilanei,C.bilocellata,C.malipoensis,C.latifolia,C.tonkinensis,C.hekouensis,C.metallica and C.laotica.[Conclusion]The results of this study not only reveal the interspecific genetic relationship of the Asian group of Caryodaphnopsis, but also have important ecological and economic significance for the study of the phylogenetic relationship and traits of Caryodaphnopsis.
