Effect of photoperiod treatment on rooting of tissue culture plantlets of Robinia pseudoacacia clonal
[Objective]The effect of different photoperiod on the occurrence of adventitious roots of black locust clones was studied,and the growth state and physiological indexes of roots at different stages under different photoperiod were analyzed,and the rooting ability was discussed to select the photoperiod that most promoted the rooting of black locust clones.[Method]Five photoperiodic treatments were applied to 4 diploid black locust clones.The rooting rate and physiological indexes of black locust were statistically analyzed under different photoperiodic conditions,and the morphological changes outside the roots were observed.At the same time,paraffin sections were taken from the base every day to observe the occurrence time of root primordia.[Result]1)Anatomical observations show that the rooting process of the tissue culture seedlings of black locust clones can be divided into three stages:callus formation stage(0-7 d),adventitious root formation stage(root induction stage)(7-10 d),adventitious root elongation stage(after 10 d).2)The results of rooting and anatomical observation showed that among the 4 black locust clones,the root primordia of D26-5 ① appeared earlier and had the strongest rooting ability(rooting rate was 92%),and the root primordia protrusion time was the earliest under 24 h photoperiod treatment,and rooting rate increased to 97.8%.3)During the rooting process,the contents of soluble sugar and chlorophyll increased with the increase of photoperiod,and the contents were the highest at 24 h photoperiod.The content of malondialdehyde(MDA)decreased,and the content of proline(PRO)increased first and then decreased,and the content of both of them was lower in 24 h photoperiod than in 16 h photoperiod.These results indicate that plants can resist environmental changes by adjusting osmotic substances in response to continuous light.[Conclusion]The ability of adventitious root generation is affected by photoperiod.The root primordium protrusion time is the earliest and the rooting ability is the strongest at 24 h photoperiod.
