4个种源无患子幼苗对低温胁迫的生理响应及抗寒性评价

[目的]通过研究不同种源无患子在低温胁迫下生理生化指标的变化,进而对不同种源无患子的抗寒性进行综合评价,筛选出较耐寒的种源,为无患子向北引种提供理论依据。[方法]采用冷库模拟低温胁迫的方法对 4 个种源的无患子幼苗进行盆栽试验,设置 0(CK)、2、4、8、12、24、48、72 h共 8 个时间梯度,分别测定其相对电导率、相对含水量、抗氧化酶系统及渗透调节物质等 11 个生理生化指标,利用各指标的隶属度值以评价不同种源无患子抗低温胁迫的能力。[结果]不同种源无患子的生理指标对低温胁迫均有不同程度的响应。相对电导率和MDA含量随着低温胁迫时间增长呈上升趋势,相对含水量呈先下降后上升的趋势,与相对含水量和MDA含量相比,叶片相对电导率对低温胁迫响应更敏感。随着低温处理时间的增长,无患子叶片中SOD、GR、POD活性和游离脯氨酸含量表现出先上升后下降的趋势,叶片CAT活性则呈先下降后上升再下降的趋势,叶片APX活性、可溶性糖和可溶性蛋白含量呈上升趋势。因此,4 个种源无患子可通过启动自身保护酶系统和渗透调节物质来增强抗寒性。[结论]根据隶属函数法计算得出低温胁迫下不同种源无患子各抗寒性相关指标的平均隶属度值,对其进行耐低温能力由强到弱排序为安徽合肥>浙江杭州>江西九江>河南新乡。
Physiological response and cold resistance evaluation of Sapindus mukorossi seedlings from four provenances to low temperature stress
[Objective]By studying the changes of physiological and biochemical indexes of Sapindus mukorossi from different provenances under low temperature stress,the cold resistance of S.mukorossi from different provenances was comprehensively evaluated,and cold-resistant provenances were selected,so as to provide theoretical basis for the introduction of S.mukorossi to the north.[Method]The cold storage simulated low temperature stress method was used to conduct pot experiment on S.mukorossi seedlings from 4 provenances,and 8 time gradient was set at 0(CK),2,4,8,12,24,48,72 h,and 11 physiological and biochemical indexes were measured,such as relative conductivity,relative water content,antioxidant enzyme system and osmotic regulatory substances.The membership value of each index was used to evaluate the ability of S.mukorossi from different provenances to resist low temperature stress.[Result]Physiological indexes of S.mukorossi from different provenances had different degrees of response to low temperature stress.The relative conductivity and MDA content increased with the increase of low temperature stress treatment time,while the relative water content decreased first and then increased.Compared with the relative water content and MDA content,the relative conductivity was more sensitive to low temperature stress.With the extension of low temperature stress time,the activities of SOD,POD,GR and free proline content of S.mukorossi leaf increased first and then decreased,the activity of CAT decreased first and then increased and then decreased,and the activity of APX,soluble sugar and soluble protein contents increased.Therefore,the S.mukorossi of 4 provenances can enhance cold resistance by activating their own protective enzyme system and osmotic regulatory substances.[Conclusion]The average membership value of cold-resistance related indexes of S.mukorossi from different provenances under low temperature stress was calculated according to membership function method.The low temperature tolerance of S.mukorossi was ranked from strong to weak as Hefei,Zhejiang,Jiujiang,Jiangxi and Xinxiang,Henan.
