Expression and Resistance Analysis of the Rice Blast Resistance Pi63 Gene Driven by Four Pi63 Promoter Constructs
Agrobacterium-mediated rice transformation was performed to obtain transgenic plants which have the the full length of rice blast resistance gene Pi63 with different cis-acting elements.The copy numbers of target gene in T1 progeny were identified by qPCR.Those T1 plants with single copy number of target gene were inoculated with blast fungus and the expression of Pi63 gene was detected before and after inoculation.The results showed that all of the expression levels of Pi63 gene with the 4 different cis-acting elements were higher after inoculation.The differential expression level indicated that cis-acting elements response to inoculation is at the region between P1 and P2 and negative elements is at the region between P0 and P1.The results may lay the foundation for revealing the molecular mechanism of rice blast resistance gene Pi63.
