正十七茶改善2型糖尿病小鼠β细胞损伤及糖脂代谢紊乱

为了探究正十七茶(ZSQT)的降血糖和降血脂作用及其机制,通过高糖高脂饲料喂养联合链脲佐菌素腹腔注射诱导建立了2型糖尿病(T2DM)模型.将造模成功的40只小鼠随机分为模型组、二甲双胍组、ZSQT高、中、低剂量组,另设8只正常对照小鼠,连续灌胃3周.每周检测体重和空腹血糖值;给药结束后进行口服葡萄糖耐量试验,计算血糖曲线面积;测定了血清中总胆固醇、甘油三酯、低密度脂蛋白、高密度脂蛋白、谷丙转氨酶和谷草转氨酶含量;通过HE染色分析了肝脏和胰腺切片组织形态;免疫荧光染色检测胰腺组织中相对α、β细胞密度.利用网络药理学预测ZSQT治疗T2DM潜在的靶点和信号通路.结果表明:ZSQT具有较好的降糖降脂作用,可以修复肝脏和胰腺损伤,增加胰腺β细胞数量,其作用机制与脂质和动脉粥样硬化等信号通路有关.
Zheng Shiqi Tea improves β Cell damage and glucose and lipid metabolism disorder in type 2 diabetic mice
To investigate the hypoglycemic and hypolipidemic effects and mechanism of Zheng Shiqi Tea(ZSQT),a type 2 diabetes(T2DM)model was induced by feeding high-sugar and fat diets and intraperitoneal injection of streptozotocin in 40 successfully modeled mice,which were randomly divided into the model group,metformin group,and high,medium,&low dose groups of ZSQT.In addition,8 normal control mice were set up and orally administered for 3 weeks.Body weight and fasting blood glucose were measured weekly,and glucose tolerance tests were performed after administration to calculate the area under the blood glucose curve.Serum total cholesterol,triglycerides,low-density lipoprotein,high-density lipoprotein,aspartate aminotransferase,and alanine aminotransferase levels were determined.The liver and pancreas tissue morphology was analyzed by hematoxylin-eosin staining,and the relative densities of α and β cells in pancreatic tissue were detected by immunofluorescence staining.Network pharmacology was used to predict potential targets and signaling pathways for T2DM treatment with ZSQT.The results show that ZSQT has a good hypoglycemic and hypolipidemic effect,can repair liver and pancreatic damage,and increase the number of pancreatic β cells,and its mechanism is related to signaling pathways such as lipid and atherosclerosis.
