含小金丸药物血清对人乳腺癌细胞株增殖、迁移和侵袭的机制研究

目的:研究含小金丸药物血清对人乳腺癌细胞株MCF-7、MDA-MB-231细胞增殖,迁移和侵袭,以及对DNMT1、DNMT3a、STAT3、β-catenin相关mRNA及蛋白表达的影响,以期探讨含小金丸药物血清对人乳腺癌细胞的作用及机制.方法:大鼠灌胃给药制备含小金丸药物血清;实验设置空白对照组和含小金丸药物血清组;以含药血清干预人乳腺癌细胞株MCF-7、MDA-MB-231;采用CCK-8法、划痕实验和Transwell侵袭实验检测细胞增殖、迁移、侵袭能力;采用 q-PCR 法检测 DNMT1、DNMT3a、STAT3、β-catenin mRNA 表达;Western Blot 法检测DNMT1、DNMT3a、STAT3、p-STAT3、β-catenin蛋白水平.结果:含小金丸药物血清呈剂量依赖性抑制乳腺癌细胞MCF-7、MDA-MB-231的增殖(P<0.05,P<0.01);抑制细胞迁移,降低划痕愈合率(P<0.05);抑制细胞的迁移和侵袭能力(P<0.01);降低细胞中DNMT1、DNMT3a、β-catenin mRNA和蛋白以及p-STAT3蛋白的表达(P<0.05,P<0.01),而对STAT3 mRNA和蛋白表达的影响无统计学差异(P>0.05).结论:含小金丸药物血清可能通过DNMT1/DNMT3a/p-STAT3/β-catenin轴抑制人乳腺癌细胞株MCF-7、MDA-MB-231细胞的增殖,迁移和侵袭的能力.
Effects of Xiaojin Pill serum on proliferation,migration and invasion of human breast cancer cell lines MCF-7 and MDA-MB-231
Objective:To study the effects of Xiaojin Pill drug-containing serum on the proliferation,migra-tion and invasion of human breast cancer cell lines MCF-7 and MDA-MB-231,as well as on the expression of DNMT1,DNMT3a,STAT3 and β-catenin related mRNAs and proteins,with a view to explore the effects and mechanisms of Xiaojin Pill drug-containing serum on human breast cancer cells.Methods:Xiaojin Pill drug-containing serum was prepared by gavage in rats.The experiments were conducted in a blank control group and a Xiaojin Pill drug-containing serum group.The drug-containing serum was used to intervene in human breast cancer cell lines MCF-7 and MDA-MB-231.The cell proliferation,migration,and invasion abili-ties were detected by CCK-8 assay,scratch assay,and Transwell invasion assay.DNMT1,DNMT3a,STAT3 andβ-catenin mRNA expressions were detected by q-PCR.The protein levels of DNMT1,DNMT3a,STAT3,p-STAT3 and β-catenin were detected by Western Blot.Results:Xiaojin Pill drug-containing serum inhibited the proliferation of breast cancer cells MCF-7 and MDA-MB-231 in a dose-dependent manner(P<0.05,P<0.01);inhibited cell migration and decreased the rate of scratch healing(P<0.05);inhibited the migration and invasive ability of the cells(P<0.01);and decreased the cellular levels of DNMT1,DNMT3a,β-catenin mRNAs and proteins as well as p-STAT3 protein expression in cells(P<0.05,P<0.01),while there was no significant difference on STAT3 mRNA and protein expression(P>0.05).Conclusion:Xiaojin Pill drug-contain-ing serum may inhibit the ability of human breast cancer cell lines MCF-7 and MDA-MB-231 cells to pro-liferate,migrate,and invade through DNMT1/DNMT3a/p-STAT3/β-catenin axis.
