Control of Coptis chinensis root rot disease by compound Trichoderma agent and its mechanism
To evaluate the effect of compound Trichoderma agent on Coptis chinensis root rot,investigate its mechanism and provide a theoretical basis for developing special microbial pesticides against this disease,compound Trichoderma agent,consisting of Trichoderma atroviride,T.longibrachiatum,T.hamatum and T.koningiopsis,and Fusarium oxysporum were applied to C.chinensis plants,respectively.The root rot symptoms were observed;the root defensive enzyme activities were determined,and fungal communities in the rhizosphere soil were analyzed by high-throughput sequencing.The results indicated that compound Trichoderma agent had obvious preventive effect on root rot caused by F.oxysporum.Compound Trichoderma agent and F.oxysporum,when applied respectively,could increase the activities of SOD,POD,CAT,PAL and PPO,inducing the plants'resistance,while strengthening effect was observed when compound Trichoderma agent and F.oxysporum were applied successively,improving plant resistance by other means.Both compound Trichoderma agent and F.oxysporum decreased the number and diversity of fungi and relative abundance of some fungi.Compared with F.oxysporum,compound Trichoderma agent had stronger fungistatic activity,inhibited pathogenic fungi such as F.oxysporum,Ilyonectria sp.,and improved soil fungal community structure.Both Trichoderma sp.and F.oxysporum could inhabit soil for a long time.Therefore,compound Trichoderma agent could prevent root rot disease caused by F.oxysporum.The disease-resistant mechanism included induced resistance,strengthening effect induced by pathogen after inoculation with compound Trichoderma agent,optimizing the fungal community,inhibiting soil pathogens,etc.Besides,compound Trichoderma agent had relatively long term of validity.In sum,compound Trichoderma agent had the potential as microbial pesticide against C.chinensis root rot disease.
