Observations on the ultrastructure of adult Gynaephora qinghaiensis antennal sensilla and tissue expression analysis of GqinOBPs
To determine the types and distribution of antennal sensilla and the function of GqinOBP genes in the process of olfactory recognition,adult antennae of Gynaephora qinghaiensis were observed using a scanning electron microscope.The expression of GqinOBP genes in the tissues of female and male adults was determined by RT-qPCR.The antennae of G.qinghaiensis exhibited smell pores and eight types of sensilla:sensilla chactica,sensilla trichodea,Böhm's bristles,uniporous peg sensilla,sensilla squamous,sensilla basiconica,sensilla cylindric,and sensilla coeloconica.Notably,sensilla chaetica,sensilla trichodea,Böhm's bristles,and sensilla basiconica had six,three,two,and three subtypes,respectively.Tissue expression analysis of GqinOBPs showed that the relative expression levels of GqinOBP 6,GqinOBP8,GqinOBP9,and GqinOBP16 in the antennae of male adults were significantly higher than in other tissues.In contrast,GqinOBP4,GqinOBP10,GqinOBP13 to GqinOBP 15,and GqinOBP17 showed highest expression levels in the wings of male adults.Furthermore,the expressions of GqinOBP2,GqinOBP12,and GqinOBP 17 were higher in female tissues,with GqinOBP17 showing a preference for females(P<0.05).This study provides foundational insights into the chemosensory mechanisms of G.qinghaiensis and offers a theoretical basis for finding molecular targets for the green control of G.qinghaiensis.
