链霉菌YC117对烟草黑胫病的防控及活性物质稳定性研究

为明确链霉菌YC117对烟草黑胫病的防控效果及活性物质的稳定性,通过形态学、生理生化特征及16S rRNA基因序列测定等方法对该菌株进行鉴定,并测定了链霉菌YC117的抑菌谱、对烟草疫霉菌丝形态的影响、发酵液的稳定性及盆栽防效.菌株YC117经鉴定确定为娄彻氏链霉菌Streptomyces rochei,具有很好的广谱抑菌活性,该菌株或其分泌物能够破坏烟草疫霉菌丝的形态结构,使菌丝出现断裂、干瘪、堆积,分枝增多和大小不一,细胞壁受损等现象.YC117菌剂在稀释100倍和200倍浓度下对烟草黑胫病的盆栽防效分别为61.54%和50.77%.发酵产生的抑菌活性物质对酸碱和热处理表现出较好的稳定性,但是对紫外照射比较敏感,紫外光照射20 min后,抑制率下降至9.52%.链霉菌YC117为防控烟草黑胫病生防制剂的开发提供了理论支持,具有较好的生防应用前景.
Control efficacy of Streptomyces YC117 against tobacco black shank disease and the stability of its active substances
To clarify the efficacy of Streptomyces YC117 in controlling tobacco black shank disease and the stability of its active substances,the strain was identified based on morphological,physiological,and biochemical characteristics,and 16S rRNA gene sequencing.The antibacterial spectrum of Streptomyces YC117 was measured,its effect on mycelium morphology of Phytophthora nicotianae was observed,and the stability of its fermentation broth was explored.In addition,its effectiveness in preventing disease in potted plants was determined.The results showed that strain YC117 was identified as Streptomyces rochei.Streptomyces YC117 exhibited broad-spectrum antibacterial activity,disrupting the morphological structure of P.nicotianae hyphae by causing fragmentation,shrinkage,increased branching,and damage to the cell wall.The potted plant experiments demonstrated control effect of 61.54%and 50.77%against tobacco black shank disease at dilutions of 100 and 200 times,respectively.The antibacterial active substances produced by fermentation showed good stability under acid and basic conditions as well as heat treatment,but they exhibited sensitivity to UV irradiation.After 20 minutes of UV irradiation,the inhibition rate decreased to 9.52%.Streptomyces YC117 holds promise for the development of biocontrol agents against tobacco black shank disease,providing foundational support for biocontrol applications.
