Isolation and identification of Ditylenchus destructor from Xinghe county,Inner Mongolia
Inner Mongolia is one of the most important potato planting area in China.Identifying the pathogen responsible for potato nematode diseases is crucial for quarantine and control in this region.Potatoes with symptoms of hollow and dry rot from Xinghe county were collected,the nematodes were separated from these potatoes,and a comprehensive analysis,including morphological identification,molecular biological identification were performed.The results showed that the nematodes share similar morphological characteristics with Ditylenchus destructor.Although there were some differences in morphological measurements,the range values were generally consistent.The rDNA-ITS sequences of these nematodes showed 100%similarity to D.destructor.Phylogenetic analysis showed that the tested population clustered into the same branch with the C-type and D-type,with sequence similarities of 99.87%and 99.26%,respectively.The symptoms caused by nematode isolates are consistent with those in the field.In summary,the nematode population in Xinghe county was identified as the C-type of D.destructor.The results providing a theoretical basis for developing control strategies for D.destructor in Xinghe county.
