为明确寄生在甘肃省天祝县草地早熟禾根部的线虫种类,本研究采用形态学和分子生物学相结合的方法,对线虫种类进行鉴定.结果表明,侵染草地早熟禾的线虫的形态学特征和形态测量值与根瘿亚粒线虫Subanguina radici-cola(Greeff,1872)Paramonov,1967 一致.rDNA-ITS和28S rDNA D2D3区域系统发育分析表明,该群体与比利时群体、中国云南群体、爱沙尼亚群体和俄罗斯群体的根瘿亚粒线虫聚为一支,且置信度较高.室内致病性试验显示,接种该线虫后,草地早熟禾表现出与田间相似的症状,繁殖系数为1.7.综上,将侵染甘肃省草地早熟禾的植物线虫鉴定为根瘿亚粒线虫,上述结果对了解粒科线虫在我国的种类多样性,及掌握该类线虫的发生、分布及潜在风险具有重要意义.
Description of Subanguina radicicola in Gansu province
This study aimed to clarify the species of plant parasitic nematodes on Poa pratensis in Tianzhu county,Gansu province.A combination of morphological and molecular methods was used to identify the species.The results showed that the morphometric and morphological characteristics of nematodes infecting P.pratensis were consistent with those of Subanguina radicicola(Greeff,1872)Paramonov,1967.The phylogenetic trees of rDNA-ITS and 28S rDNA D2D3 regions indicated that the population was clustered with S.radicicola population from Belgian,Yunnan,Estonian,and Russian,with high confidence.Meanwhile,laboratory pathogenicity test showed that all plants inoculated with nematodes showed similar symptoms observed in the field,and reproduction factor was 1.7.Therefore,the nematode infecting P.pratensis in Gansu was identified asS.radicicola,which is of great significance for understanding the biodiversity of Anguinidae nematodes in China and grasping the occurrence,distribution and potential risks of such nematodes.
