Species identification of root-knot nematodes infecting Cucumis melo var.saccharinus in Hainan province
In order to explore the occurrence of the root-knot nematode disease of Cucumis melo var.saccharinus and understand the species of nematode in Hainan province,the disease occurrence was investigated and the pathogens from samples randomly collected from the main producing area in Hainan in April,2023 were identified using morphology and molecular biology methods.The results showed that the root-knot nematode disease occurred in all the 11 investigated places,and nematode disease was most severe at sampling point 4-1 in Haorui village,Dongfang city,with root-knot grade of five.Meloidogyne incognita,M.arenaria and M.enterolobii were identified based on morphological characteristics of second-stage juveniles and female perineal pattern in combination with 28S rDNA D2D3 and SCAR sequences.M.incognita was the most widespread and was detected at eight sampling sites.Mixed populations of M.incognita and M.enterolobii or M.incognita and M.arenaria were happened.The above results indicate that M.incognita is the dominant population of root-knot nematode,which will provide a theoretical basis for the effective control of root-knot nematodes on Cucumis melo var.saccharinus in Hainan province.
