The correlations between the resistance of common potato varieties to Globodera rostochiensis and the H1 gene in Yunnan province
To identify the differences in resistance among common potato varieties to Globodera rostochiensis in Yunnan province,25 varieties, including both common and widely cultivated ones, were selected for evaluation using Koroleva's method. In this study the molecular markers N146,N195,57R,TG432,Gro1-4-1,and the H1 resistance gene were employed to evaluate resistance levels and analyze the correlation between resistance and H1 gene expression. The results showed that among the 25 varieties tested, seven varieties exhibited resistance, with less than five cysts or females of G.rostochiensis on their root systems, while the remaining varieties, with more than five cysts or females, were susceptible.Molecular marker analysis revealed that ten varieties containing the G.rostochiensis resistance gene H1.Further investigation into the relative expression of the H1 gene after inoculation with G.rostochiensis showed that in seven varieties,namely'V7''Huangjinshu''Huishu 19''Xuanshu 5''Yunshu 202''Yunshu 304',and'Yunshu 505',the H1 gene expression was up-regulated compared to uninoculated controls. However,in three varieties'P14187''Huishu 15'and'Xisen 3',the H1 gene expression was down-regulated.The study concluded that the resistance of potato varieties carrying the H1 gene were positively correlated with H1 gene expression. These findings provide foundation for selecting and applying potato varieties resistant to G.rostochiensis and for further elucidating the molecular mechanisms underlying potato resistance to G.rostochiensis.
