牛膝枯萎病病原鉴定、生物学特性及防治药剂筛选

以河南省焦作市牛膝枯萎病样品为试材,采用组织分离培养法对其病原进行分离纯化,利用蘸根接种法对分离物的致病性进行测定.通过形态学、ITS序列和多基因(EF1-α、Tub、RPB2和PRO1/2等)序列分析方法完成了牛膝枯萎病病原菌鉴定,并对病原菌的生物学特性进行了研究.结果表明,牛膝枯萎病的病原菌为层出镰刀菌(Fusarium proliferatum).该病菌可在多种培养基上生长,在燕麦琼脂培养基上生长最快;最适生长温度为28 ℃,最适pH值为7.0,在全黑暗培养条件下生长速率最高;可利用多种碳、氮源,其中以蔗糖为碳源和蛋白胨为氮源的培养条件最利于菌丝的生长.毒力测定结果表明,选用的6种杀菌剂中戊唑醇和咯菌腈的抑菌效果最好,EC50值分别为3.03和2.36 mg·L-1.本研究是F.proliferatum引起牛膝枯萎病的首次报道,研究结果可为牛膝枯萎病病害的田间诊断和科学防治提供依据.
Isolation,biological characterization of the pathogen causing Fusarium wilt on Achyranthes bidentata and fungicide screening
The diseased plants of Achyranthes bidentata showing typical symptoms of Fusarium wilt were col-lected from Jiaozuo,Henan Province,China.Tissue separation method was used to obtain potential pathogenic fungal isolates,and the pathogenicity of these isolates was determined by root-dipping inoculation method.Fu-sarium proliferatum was identified as the causal agent of the disease based on morphological characteristics,ITS sequence and polygenic analysis(EF1-α,Tub,RPB2 and PRO1/2).F.proliferatum isolates showed maximum radial growth at 28 ℃ and pH 7.0 on oatmeal agar medium under dark conditions.The pathogen could utilize multiple carbon and nitrogen sources,with the best carbon and nitrogen sources of sucrose and peptone,respec-tively.To our knowledge,this is the first report of F.proliferatum causing Fusarium wilt on A.bidentata.The results provide a scientific basis for diagnosis and control of Fusarium wilt of A.bidentata.Toxicity test of the six fungicides on F.proliferatum showed that these fungicides had certain inhibitory effects on the pathogen,and tebuconazole and fludioxonil exhibited relative higher inhibitory effects,with EC50 values of 3.03 and 2.36 mg· L-1,respectively.
