Preliminary study on the composition of the fungal and bacterial community and potential pathogen carried by the imported Hippeastrum vittatum bulb from Peru
The aim of this study was to investigate the composition of the fungi and bacteria,and the potential pathogens in the imported Hippeastrum vittatum bulb samples from Peru.Five batches of imported H.vittatum bulb samples were collected from Ningbo Port.Five root samples and five top samples of H.vittatum were obtained,respectively.Bioinformatics method used Pacbio amplicon sequencing technology based on 16S rRNA gene and ITS,and traditional isolation and culture method were used to determine and compare the composition of the fungal and bacterial flora and potential pathogens in the samples.The analysis of fungal flora composition in the root and top of the bulb showed that Ascomy-cota had the highest fungal abundance at the phylum level.At the genus level,Penicillium was the most abundant fungal flora,with an average relative abundance of 68.73% and 26.20% at the root and top of the bulb,respectively.At the species level,Penicillium brevicompactum was the most abundant fungal species with an average relative abundance of 58.54% and 24.02%,respectively.The results of bacterial flora composition analysis showed that Proteobacteria had the highest bacterial abundance at the phylum level.At the genus level,Pantoea was the most abundant bacterial flora,with an average relative abun-dance of 66.55% and 74.50% at the root and top of the bulb,respectively.At the species level,Pantoea agglomerans was the most abundant bacterial species,with an average relative abundance of 52.18% and 66.50%,respectively.The results of amplicon sequencing revealed that Burkholderia gladioli might be car-ried in the roots of the imported H.vittatum bulbs from Peru.The pathogen B.gladioli pv.alliicola is listed in Chinese quarantine pest.A variety of pathogens were obtained by isolation and culture method,such as B.gladioli.There were differences in the dominant genera of the flora composition between the root and the top of the imported H.vittatum bulbs.The amplicon sequencing method could provide infor-mation for screening out potential quarantine pathogen.This study provides a new reference for detection and identification of quarantine pathogen in port.
