基于RPA的菲利普木蠹象分子鉴定技术

为建立快速、灵敏检测菲利普木蠹象的重组酶聚合酶扩增检测(RPA)方法,本研究利用菲利普木蠹象的COI基因序列,设计筛选特异性引物PH3,并对引物特异性和灵敏度进行检测.结果表明,设计的引物能够特异检测出菲利普木蠹象,该引物检测菲利普木蠹象的灵敏度为0.045 ng/μL.所建立的RPA方法特异性强、灵敏度高,可用于口岸一线对菲利普木蠹象的检测.
Molecular identification of Pissodes piniphilus(Herbst)based on RPA
To establish a rapid and sensitive recombinase polymerase amplification assay(RPA)method for the detection of Pissodes piniphilus.This study designed and screened specific primers PH3 based on the COI gene sequence of Pissodes piniphilus,and detected the specificity and sensitivity of the primers.The results showed that the designed primer was able to specifically detect Pissodes piniphilus,with a sensitivity of 0.045 ng/μL.The established recombinase polymerase amplification assay(RPA)method has strong specificity and high sensitivity for detecting Pissodes piniphilus at port.
