Evaluation of Antibacterial Activity and Biocontrol Potential of Eight Strains of Endophytic Fungi from Colletotrichum siamense
In order to explore the inhibition effect and biocontrol potential of endophytic fungi strains against grape anthracnose,eight endophytic fungi strains isolated from healthy Sophora alopecia plants were antagonistic,and Colletotrichum siamense was used as the target bacteria.The antibacterial activity of endophytic fungi against Colletotrichum siamense was investigated by the method of in-dish standoff,mycelium growth rate,spore germination and in vitro control test.The results indicated that each of the eight strains of endophytic fungi found in bitter beans exhibited a noteworthy inhibitory impact on the Cs-2-7-NX strain.Among them,strain HD31 had the best comprehensive performance among the three treatments,and the inhibition rate of strain HD31 after 3 days of early inoculation and then inoculation of target bacteria could reach 84.62%.Optical microscopic observation revealed that strain HD31 had the capability to induce hyphal rupture and node enlargement.The highest inhibition rate of 80.25%against the mycelial growth of Cs-2-7-NX strain was observed when using the fermentation filtrate of strain HD35 at a concentration of 25%.the fermentation filtrate of strain HD31 showed the highest inhibition rate of 72.65%.At a concentration of 40%in the fermentation filtrate,the HD31 strain had the highest inhibition rate of 83.07%on the germination of Cs-2-7-NX strain spores.Additionally,the volatile material produced by the HD31 strain at 6 days pre-inoculation demonstrated the highest inhibition rate of 82.09%on the Cs-2-7-NX strain.The results of in vitro control effect test indicatedthat the protective efficacy of the fermentation filtrate of antagonistic endophytic fungi,when applied prior to inoculation with the pathogenic fungi Cs-2-7-NX strain was generally higher than that of the fermentation filtrate of antagonistic endophytic fungi sprayed after 3 days of inoculation.Among the various samples tested,it was observed that the culture filtrate containing strain HD31 at a concentration of 10%exhibited the most effective preventive action against the Cs-11 strain,with a control efficacy of 85.59%,which suggested that strain HD 13 exhibits promising potential as an initial bacterium for conducting research on the development of environmentally friendly strategies to control grapevine anthracnose.
