摘要

刺葡萄原产中国,是东亚种群的特色野生资源之一,通过SSR分子标记分析刺葡萄的遗传多样性,对其物候期、植物学特性和生根特性等进行评价.本研究共筛选出8份浙江特异种质资源标记为VD1、VD3、VD5、VD6、VD9、VD11、VD12和VD15,聚类分析显示刺葡萄有着很强的地域性;刺葡萄生根困难,在无任何加温等措施下,VD15生根率最高,为13.33%,愈伤组织诱导率为86.67%;切片结果表明,皮刺的位置表皮细胞延伸增多,葡萄愈伤组织形成与生根是两个独立的过程.以上结果为刺葡萄资源的筛选和利用奠定了基础.

Abstract

Vitis davidii is one of the characteristic wild grape resources of East Asian populations,originating from China.The genetic diversity was analyzed,using SSR molecular markers,and their phenological,botanical,and rooting characteristics were evaluated.This study screened 8 specific germplasm resources in Zhejiang region,including VD1,VD3,VD5,VD6,VD9,VD11,VD12,and VD15.Cluster analysis showed that Vitis davidii have strong regional characteristics.The rooting of Vitis davidii was difficult,without any heating or other measures,VD15 had the best cutting rooting,which had 13.33% of rooting rate,and 86.67% of callus induction rate.The slicing results showed that the epidermal cells at the location of the prickles extended more.The formation of grape callus tissue and rooting were two independent processes.The above results have laid the foundation for the screening and utilization of Vitis davidii resources.
