番茄不孕病毒(TAV)脱除对‘怀白菊’活性氧代谢的影响

以前期获得的脱除番茄不孕病毒(TAV)的‘怀白菊’试管苗为试材,从同工酶谱和DNA分子简单序列间重复(ISSR)多态性方面分析其遗传稳定性,在此基础上,对其继代培养过程中叶片中的活性氧代谢进行了研究。结果表明, TAV脱除没有改变‘怀白菊’叶片中SOD、POD同工酶谱带及ISSR扩增条带的数目,但相应同工酶谱带亮度增强;也没有改变‘怀白菊’试管苗继代培养过程中膜脂过氧化相关指标(O·2ˉ生成速率及H2O2和MDA含量)和保护系统相关指标(保护酶SOD、POD、CAT、APX和GR,抗氧化物质GSH和AsA)的变化趋势,但却显著降低了相同时间点膜脂过氧化相关指标的绝对值,降幅最大值超过30%(O·2ˉ生成速率、MDA含量),显著增加了保护酶活性及抗氧化物质的含量,增幅最大值超过了50%(APX活力、GSH含量)。以上说明, TAV脱除不仅没有改变‘怀白菊’的遗传稳定性,而且降低了其活性氧代谢水平,提高了其抗氧化能力。
Effect of TAV-Free on Active Oxygen Metabolism ofChrysanthemum morifolium‘Huaibai’
Tomato aspermy virus (TAV)-freeChrysanthemum morifolium‘Huaibai’ test-tube plantlets were used as experimental materials. In this study, genetic stability of TAV-free ‘Huaibai’ test-tube plantlets was ana-lyzed from isozyme zymogram and DNA molecular ISSR polymorphism. Based on this, active oxygen metabo-lism of TAV-free ‘Huaibai’ plantlets had been studied during subculture. The results showed that: TAV-free didn’t change the number of SOD isozyme and POD isozyme bands but the band brightness were brighter than the virus-carrying plantlets, and also didn’t change the number and brightness of ISSR ampliifcation bands. So TAV-free maintained its genetic stability and increased the activities of SOD and POD. TAV-free also didn’t change the trend of lipid peroxidation related indexes (O·2ˉproduce rate, the MDA and H2O2 contents) and pro-tection system related indexes (protective enzymes SOD, POD, CAT, APX and GR activities; antioxidants GSH and AsA contents) during subculture, but signiifcantly reduced the absolute value of lipid peroxidation related indexes and signiifcantly increased activities of protective enzyme and the contents of antioxidants at the same time of subculture. The maximum decrease of more than 30% (O·2ˉ produce rate and MDA content) and the max-imum increase of more than 50% (APX activity and GSH content) were gotten in the leaves of TAV-free plant-lets. In conclusion, TAV-free didn’t only change genetic stability ofChrysanthemum morifolium ‘Huaibai’, but also reduced the active oxygen metabolism and improved its antioxidant capacity.
