Construction of in vitro rapid propagation technology system of Melliodendron xylocarpum
Melliodendron xylocarpum,a deciduous tree endemic to China,possesses exceptional ornamen-tal,oil,medicinal and timber value,but its genetic resources are currently endangered.This study em-ployed young stem segments of M.xylocarpum as the primary material and conducted research across four phases including initiation culture,multiplication culture,rooting culture and transplanting in order to establish an in vitro rapid propagation technology system of M.xylocarpum.The results showed that the combination of 75%ethanol immersion for 30 s and 0.1%HgCl2 immersion for 12 min at the initiation cul-ture stage had the lowest contamination rates with fungal contamination rate of 11.11%and bacterial con-tamination rate of 2.22%,and the highest induction rate was 69.48%and survival rate was 80.00%,which were able to effectively sterilize the surface of M.xylocarpum stem segments explants.During the multipli-cation culture stage,1.0-1.5 mg·L-1 BA significantly promoted the growth of in vitro shoots with the high-est multiplication rate of 4.53-4.70.Additionally,2.0 mg·L-1 IAA was effective in promoting root induction of in vitro shoots with rooting rate of 87.23%and had the highest transplanting survival rate of 88.33%with 77.05%seedling rate in the rooting culture phase.This study has pioneered a highly efficient in vitro rapid propagation technology for M.xylocarpum,providing crucial technical support for its large-scale propaga-tion and industrialization promotion.
