Identification and expression analysis of GeBP gene family in red raspberry
Glabrous-enbancer binding protein(GeBP)is a unique protein in plants, which plays an important role in plant growth and development, hormone levels and stress. In this study, using'Heritage'red rasp-berries as materials, the GeBP gene family members were identified and their expression patterns were studied by transcriptome sequencing, bioinformatics analysis and qRT-PCR.The results showed that:three members of the GeBP gene family the protein amino acid number ranged from 236-407,the molecular weight ranged from 26.66-42.25 kDa,and the isoelectric point ranged from 4.84-9.52.RuGeBP-1 and Ru-GeBP-2 are unstable proteins, and RuGeBP-3 is stable. All three are hydrophilic proteins, contain most ami-no acids as serine, have no signal peptide, are non-secreted proteins, are localized in the nucleus, are non-transmembrane proteins, contain DUF573 conserved domain, secondary structure prediction showed that the proteins encoded by RuGeBPs were composed of α helix,β corner, random curling and folding ex-tension chains, and the tertiary structure prediction results were consistent with the secondary structure. The analysis of gene structure and conserved motifs showed that RuGeBP gene family had 0-2 introns, and have six important conserved motifs;Phylogenetic analysis showed that red raspberry and strawberry were more closely related. The promoter cis-acting elements indicated that RuGeBP gene family was mainly in-volved in hormone response, defense and stress response, plant growth and development.RuGeBP gene family had tissue-specific expression, which was mainly expressed in early fruit development and roots.Ru-GeBP-1,RuGeBP-3 might be related to the response mechanism of abscisic acidand other hormones in fruit and the growth a of roots,RuGeBP-2 may be involved in root growth and defense mechanisms.
