BEL1(homeodomain transcription factor BELL 1)是普遍存在于植物体内并发挥调控生殖和形态等发育作用的转录因子.本文以蒺藜苜蓿BEL1为研究对象,通过亚细胞定位、生物信息学分析、表达模式分析、酵母表达和原核表达实验初步探究MtBEL1功能.结果表明,MtBEL1完整编码区为2 517 bp,编码839个氨基酸.对蒺藜苜蓿BEL1转录因子上游区域进行顺式作用元件预测分析表明,MtBEL1基因可能在蒺藜苜蓿植物生长调节物质信号通路中发挥作用.进化树分析显示MtBEL1蛋白与蚕豆和豌豆同源蛋白亲缘关系最近.该基因在根部表达最为活跃,叶中表达量最低,MtBEL1基因的表达受6-BA、ABA、GA和IAA的促进,MeJA的抑制.亚细胞定位分析显示该蛋白主要在细胞核和细胞质中发挥作用.酵母表达分析表明:MtBEL1蛋白不具有毒性,存在转录自激活活性.原核表达分析表明该蛋白为可溶性蛋白.
Expression,subcellular localization and transcriptional self-activation analysis of transcription factor BEL1 in Medicago truncatula
BEL1(Homeodomain transcription factor BELL 1)is a transcription factor commonly found in plants and plays a role in regulating reproduction and morphological development.Taking Medicago trun-catula BEL1 as the research object,the function of MtBEL1 was preliminarily explored through subcellular localization,bioinformatics analysis,expression pattern analysis,yeast expression and prokaryotic expres-sion experiments.The results showed that the complete coding region of MtBEL1 was 2 517 bp and encoded 839 amino acids.The cis-acting element prediction analysis of the upstream region of the transcription fac-tor BEL1 showed that the MtBEL1 gene may play a role in the signaling pathway of plant growth regulators in M.truncatula.Phylogenetic tree analysis showed that BEL1 protein was closely related to homologous pro-teins of broad bean and pea.The expression of MtBEL1 gene was the most active in root,and the lowest in leaf,and the expression of this gene was promoted by 6-BA,ABA,GA,IAA,and inhibited by MeJA.Subcellular localization analysis showed that the protein mainly plays a role in the nucleus and cytoplasm,and the ex-pression analysis of MtBEL1 in yeast showed that MtBEL1 protein was not toxic and had transcriptional self-activating activity.Prokaryotic expression analysis showed that the protein was soluble protein.
