Physiological and transcriptomic insights into leaf color variation in half-sib progeny of Koelreuteria paniculata'Xinye'
The half-sib progenie Koelreuteria paniculata'Xinye'with yellow and green leaves were used as material.The physiological indexes of the plant materials sampled in July and October were measured,in-cluding color variation,photosynthesis indicators,chlorophyll fluorescence parameters,etc.Moreover,RNA-seq and RT-qPCR assays were also performed.The results show that the pigment contents of all green-leaf seedlings were nearly twice as high as those of yellow-leaf seedlings.Additionally,only two in-dexes(i.e.,Ci and Fv/Fm)were lower in green-leaf seedlings than in the yellow-leaf seedlings,while all other physiological indexes were higher in green-leaf seedlings.Furthermore,compared to the plant materials sampled in July,the pigment and starch contents and photosynthesis ability were down-regulated,where-as the soluble sugar content and Fv/Fm were up-regulated in the plant materials sampled in October.Tran-scriptome results suggest that the yellow leaves of the seedlings may be attributed to the down-regulation of genes associated with pathways such as chlorophyll biosynthesis,chloroplast development,and flavo-noid biosynthesis.RT-qPCR assay further confirmed the reliability of the RNA-seq data.This work provides insight into further investigating the mechanisms of the leaf color formation of the plants as well as creat-ing new germplasm with colorful foliage.
