Tissue culture of an endangered mangrove plant-Lumnitzera littorea
Lumnitzera littorea is difficult to reproduce naturally,and the wild population is on the verge of extinction.In order to protect the germplasm resources,this study investigated the sterilization effects of different NaClO solution concentrations and sterilization time on seedlings leaves,light intensities and plant growth regulator concentrations on callus induction and proliferation of L.littorea based on MS medi-um.The results showed that:50 g·L-1 NaClO sterilized for 10 min had the best effect,which decreased contamination rate and severe browning rate of explants to 15.5%and 35.5%,respectively.The callus in-duction effect of the explants in the dark was the best,and the callus induction rate was 19.4%.Before in-oculation,the explants of the middle part leaf were hurt and immersed in 0.1 g·L-1 NAA+0.05 g·L-1 6-BA solution for 30-40 min,and then inoculated in 1.2 mg·L-1 NAA+0.1 mg·L-1 6-BA medium cultured in the dark for 30 days which caused rate of the callus induction up to 50.0%.1.2 mg·L-1 IAA+0.1 mg·L-16-BA and 1.0 mg·L-1 6-BA+0.1 mg·L-1 NAA were the best medium for callu proliferation,and the dry and fresh weight of explants reached the maximum.
