Effect of Shoutai Pill on Aerobic Glycolysis Inhibition in Decidual Tissue of Recurrent Spontaneous Abortion Mice
Objective To observe the effect of Shoutai Pill on the prevention and treatment of recurrent spontaneous abortion(RSA),and to study its effects of regulating aerobic glycolysis.Methods Forty CBA/J mice were randomly divided into normal group,RSA group,dydrogesterone group,and Shoutai Pill group,10 mice in each group.The mice were administered with distilled water,dydrogesterone solubilizer,and Shoutai Pill by gastrogavage for 14 successive days,respectively.After 14 days,female CBA/J rats in normal group and male BALB/c rats in BALB/c group were mated at 2∶1,and female rats in other 3 groups were mated with male DBA/2 rats at 2∶1.RSA model was established,and gastrogavage was continued until the 10th day of embolization according to previous methods.The rate of mouse embryo loss in each group was calculated.The decidua tissue was observed by HE staining.The expressions of hexokinase(HK)2,M2 pyruvate kinase(PKM2),lactate dehydrogenase(LDH)A,B-cell lymphoma-2(Bcl-2),B-cell lymphoma-2 associated X protein(Bax),and cysteinyl aspartate specific protease-3(Caspase-3)in the decidua tissue of mice were detected by Western Blot and Real-time fluorescence quantitative PCR(qRT-PCR).Lactic acid(LA)content was detected by lactate dehydrogenase colorimetry.The activities of HK,PK,and LDH were also detected.Results Compared with the normal group,the embryo loss rate of mice in RSA group increased(P<0.05),decidua tissue cells were reduced with loose arrangement,cytoplasm vacuolated,and partial nuclei disappeared.And the protein and mRNA expressions of HK2,PKM2,LDHA,and Bcl-2 decreased(P<0.05).The protein and mRNA expressions of Bax and Caspase-3 increased(P<0.05).And the activities of HK,PK,and LDH decreased(P<0.05),and LA content lowered(P<0.01).Compared with RSA group,the embryo loss rate of mice in dydrogesterone group and Shoutai Pill group decreased(P<0.05).The arrangement of decidual tissue cells was relatively dense,with cytoplasmic edema in few decidual cells,cytoplasm vacuolated,and richer cytoplasm.And the protein and mRNA expressions of HK2,PKM2,LDHA,and Bcl-2 in the decidual tissue of mice increased(P<0.05).The protein and mRNA expressions of Bax and Caspase-3 decreased(P<0.01).The activities of HK,PK,and LDH increased(P<0.05),and LA content increased(P<0.01).Conclusion Shoutai Pill played roles in preventing and treating RSA possibly through regulating the level of aerobic glycolysis in decidua tissue,elevating the content of lactic acid,reducing the apoptosis of decidua cells,and further reducing the embryo loss rate.
