Effects of Guben Qushi Huayu Formula on the Skin Lesions and Spleen Water Transport Related Substances in a Mouse Model of Psoriasis Recurrence
To study the possible mechanism of Guben Qushi Huayu Formula(GQHF)in treating and postponing the relapse of psoriasis.Methods Totally 42 Balb/c mice were randomly divided into normal group(N),model group(M),relapse model group(M+M),methotrexate group(MTX),and high/medium/low dose GQHF groups(GQHF-H/M/L),6 mice in each group.Except Group N,mice in all the other medication groups were administered by corresponding medicines by gastrogavage from the first day.GQHF-H/M/L groups were administered with 22.75,11.34,and 5.69 g/kg GQHF daily respectively.MTX at 1 mg/kg per day was administered to mice in Group MTX by gastrogavage.Drinking water at 10 mL/kg was administered to mice in Group N and Group M.All medication lasted for 15 successive days.On the 4th day,except Group N and M,a psoriasis-like skin lesion model was established in mice of the rest groups by applying 50 mg 5%imiquimod cream.On the 38th day,except Group N,a psoriasis-like skin lesion relapse model was established in mice of the rest groups by applying imiquimod cream.Psoriasis area and severity index(PASI)scores of psoriasis were evaluated,and the activities of adenosine triphosphatase(ATPase)and phase Ⅱ metabolic enzymes(SULT1A1,UGT1,GST,SULT1 E1),gene and protein expressions of Aquaporin(AQP)3 in spleen of mice in each group were detected.Results Compared with Group M+M,skin lesions were significantly reduced in GQHF groups and Group MTX(P<0.05,P<0.01).Compared with Group N,Na+-K+-ATPase,Ca2+-Mg2+-ATPase,and total ATPase(T-ATPase)activities in spleen significantly decreased(P<0.05)in Group M and Group M+M.The gene and protein expressions of AQP3 increased significantly(P<0.05),and there was no statistical difference in phase Ⅱmetabolic enzyme activities(P>0.05).Compared with Group M+M,Na+-K+-ATPase,Ca2+-Mg2+-ATPase,and T-ATPase activities significantly increased in Group GQHF-H and Group MTX(P<0.05).AQP3 gene and protein expressions in spleen significantly decreased(P<0.01).Group GQHF-H showed better efficacy than Group MTX in preventing relapse and reducing AQP3 protein expression(P<0.05).Conclusion GQHF achieved therapeutic effects in treating and postponing the relapse of psoriasis possibly by elevating the activity of ATPase in spleen,reducing the expression of AQP3 protein,and improving the local internal humid environment.
