仙甲汤抑制IL-6/JAK2/STAT3信号通路改善大鼠前列腺增生

目的 基于白细胞介素(IL)-6介导的炎症反应,探讨仙甲汤改善大鼠前列腺增生(BPH)的作用及机制。方法 将36只雄性SD大鼠随机分为空白组、模型组、仙甲汤高、中、低剂量组、非那雄胺组,每组6只。除空白组外,每组皮下注射丙酸睾酮(5 mg/kg)连续30天制备BPH模型。造模完成后,仙甲汤高、中、低剂量组分别给予仙甲汤27。18、13。59、6。80 g/kg灌胃,非那雄胺组给予非那雄胺0。45 mg/kg灌胃,空白组和模型组给予等体积生理盐水,连续30天。测量大鼠体重、前列腺湿重及体积并计算前列腺指数;HE染色法观察前列腺组织病理学变化;甲苯胺蓝染色法观察前列腺组织肥大细胞并计数;ELISA法检测前列腺组织IL-6、IL-10含量;Western Blot检测前列腺组织酪氨酸蛋白激酶2(JAK2)、磷酸化 JAK2(p-JAK2)、转录激活因子 3(STAT3)、磷酸化 STAT3(p-STAT3)表达;Real-time PCR检测前列腺组织IL-6、JAK2、STAT3 mRNA表达。结果 空白组大鼠前列腺组织中腺体饱满、规则;模型组大鼠前列腺腺体不规则,腺上皮细胞增生,基质内出现纤维组织增生,并有炎性细胞浸润;干预组出现不同程度好转。与空白组比较,模型组大鼠前列腺体积、湿重、指数、肥大细胞数、IL-6含量、p-STAT3、p-JAK2蛋白及IL-6、JAK2、STAT3 mRNA表达升高,前列腺组织IL-10含量降低(P<0。01)。与模型组比较,仙甲汤高、中剂量组和非那雄胺组前列腺体积、湿重、指数、肥大细胞数,IL-6含量、p-STAT3、p-JAK2蛋白及IL-6、JAK2、STAT3 mRNA表达降低,前列腺组织IL-10含量升高(P<0。05,P<0。01);仙甲汤低剂量组大鼠前列腺体积、湿重、STAT3 mRNA表达下降(P<0。05,P<0。01)。与非那雄胺组比较,仙甲汤高剂量组大鼠前列腺体积、湿重、指数、肥大细胞数、IL-6含量、p-STAT3、p-JAK2蛋白表达降低,前列腺组织IL-10含量升高(P<0。05,P<0。01)。结论 仙甲汤能显著改善大鼠BPH,其机制可能与抑制IL-6/JAK2/STAT3通路过度激活有关。
Xianjia Decoction Improves Prostatic Hyperplasia in Rats Via Inhibiting IL-6/JAK2/STAT3 Signaling Pathway
Objective To investigate the effect and mechanism of Xianjia Decoction(XD)on prostatic hyperplasia(BPH)in rats based on IL-6 mediated inflammatory response.Methods Totally 36 male SD rats were randomly divided into blank group,model group,XD high-dose,medium-dose,low-dose group and finasteride group,with 6 rats in each group.Except the blank group,each group was subcutaneously injected with testosterone propionate(5 mg/kg)for 30 consecutive days to prepare BPH model.After modeling,XD high,medium and low dose group were intragastric administration XD 27.18,13.59 and 6.80 g/kg,respectively,while finasteride group was intragastric administration finasteride 45 mg/kg,the blank group and model group were intragastric administration equal volume normal saline,the course was 30 consecutive days.Body weight,wet prostate weight and prostate volume of rats were measured and prostate index was calculated.HE staining was used to observe the histopathological changes of prostate.Mast cells of prostate tissue were observed and counted by toluidine blue staining.lnterleukin-6(IL-6)and interleukin-10(IL-10)were detected by ELISA.Janus kinase 2(JAK2),phosphorylated JAK2(p-JAK2),signal transducer and activator of transcription 3(STAT3)and phosphorylated STAT3(p-STAT3)were detected by Western Blot.The mRNA of IL-6,JAK2 and STAT3 in prostate tissue was determined by Real-time PCR.Results The glands in the prostate tissue of the blank group rats full and regular,while it is irregular,with proliferation of glandular epithelial cells,fibrous tissue proliferation in the matrix,and infiltration of inflammatory cells in the prostate gland of the model group rats.The intervention group showed varying degrees of improvement.Compared with the blank group,the prostate volume,wet weight,prostate index,mast cell number,IL-6 content,p-STAT3,p-JAK2 protein and IL-6,JAK2,STAT3 mRNA expression increased,and IL-10 content decreased in the model group(P<0.01).Compared with the model group,the volume,wet weight,prostate index,the number of mast cells,the content of IL-6,p-STAT3 and p-JAK2 protein and IL-6,JAK2 and STAT3 mRNA expression decreased,and the content of IL-10 in prostate tissue was increased in the high and medium-dose XD group and finasteride group(P<0.05,P<0.01).The prostate volume,wet weight and STAT3 mRNA expression of rats in low dose XD group decreased(P<0.05,P<0.01).Compared with the finasteride group,the prostate volume,wet weight,index,mast cell number,IL-6 content,p-STAT3 and p-JAK2 protein expression decreased,and IL-10 content increased in the high-dose XD group(P<0.05,P<0.01).Conclusions XD can significantly improve BPH in rats,and its mechanism may be related to inhibiting the overactivation of IL-6/JAK2/STAT3 pathway.
