解毒祛瘀滋肾方对强的松所致MRL/lpr小鼠脂质代谢紊乱的调控作用

目的 研究糖皮质激素对系统性红斑狼疮(SLE)模型小鼠脂质代谢的影响,以及解毒祛瘀滋肾方对脂质的调节作用。方法 12只MRL/lpr小鼠按随机数字表法分为模型组、西药组(10 mg/kg强的松)和中西药组(10 mg/kg强的松+20 g生药/kg解毒祛瘀滋肾方),每组4只。分别于给药后10、30天采血。采用鸟枪法脂质组学技术分析血清脂质组,经方差归一化法标度后进行主成分分析,进一步应用非参数检验筛选在组间存在显著性差异的脂质分子。结果 与模型组比较,10 mg/kg强的松干预10天和30天,脂质谱均发生了显著变化,强的松干预10天后,MRL/lpr小鼠血清中24种脂质与模型组差异有统计学意义(P<0。05),干预30天后,影响的脂质种类更多(51种)。解毒祛瘀滋肾方联合强的松干预10天后,16/24种脂质恢复到模型组的水平,联合干预30天后,25/51种脂质恢复到模型组的水平。结论 强的松会干扰MRL/lpr小鼠脂质代谢,解毒祛瘀滋肾方对脂质代谢紊乱有显著的调节作用,可能是解其减轻糖皮质激素不良反应的机制之一。
Regulatory Effects of Jiedu Quyu Zishen Recipe on Prednisone-Induced Lipid Metabolism Disorders in MRL/lpr Mice
Objective investigate the impact of glucocorticoids on lipid metabolism in a systemic lupus erythematosus(SLE)mouse model,and the regulatory effect of Jiedu Quyu Zishen Recipe(JQZR)on lipids.Methods Totally 12 MRL/lpr mice were randomly divided into the model group,Western medicine group(10 mg/kg prednisone),and combined therapy group(10 mg/kg prednisone+20 g/kg JQZR),with 4 mice in each group.Blood samples were collected at 10 and 30 days post-treatment.Serum lipid profiles were analyzed using Shotgun technique,followed by principal component analysis after unit variance scaling.Subsequently,non-parametric tests were applied to identify lipids with significant differences among groups.Results Compared with the model group,treatment with 10 mg/kg prednisone for 10 and 30 days led to significant changes in the lipid profile.After 10 days of prednisone treatment,24 kinds of lipids in the serum of MRL/lpr mice showed significant differences(P<0.05),and after 30 days of treatment,more lipids(51 kinds)were affected.After 10 days of combined treatment with JQZR and prednisone,16 out of 24 lipids returned to the level of the model group.Following 30 days of combined treatment,25 out of 51 lipids returned to the level of the model group.Conclusion Prednisone can interfere with lipid metabolism in MRL/lpr mice,and JQZR has a significant regulatory effect on lipid disorders,which may be one of the mechanisms of alleviating the side effects of glucocorticoids.
