化湿通痹方对勃起功能障碍大鼠血脂水平、血管内皮细胞与肠道菌群的影响

目的 观察化湿通痹方(HTF)对勃起功能障碍大鼠血脂水平、血管内皮细胞功能及肠道菌群的影响.方法 将SD大鼠随机分为空白组、假手术组、模型组、西地那非组、HTF低、中、高剂量组,每组7只.采用高脂血症联合双侧髂内动脉结扎造模,造模后灌胃给药,西地那非组予西地那非(4.5mg/kg)灌胃,HTF组分别予HTF低、中、高剂量(3.24、6.48、12.96 g/kg)灌胃,其他三组予等量生理盐水灌胃.干预6周后,采用多道生理信号记录仪测量阴茎海绵体内压(ICP)、平均颈动脉压(MAP),计算ICP/MAP,记录大鼠勃起次数;采用全自动生化仪检测血清总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平,ELISA法检测大鼠血清血管内皮生长因子(VEGF)、内皮素1(ET-1),NO微板法检测海绵体组织一氧化氮(NO)水平.16 SrDNA测序检测大鼠肠道菌群.结果 与本组治疗前比较,HTF各剂量组大鼠血清LDL-C水平降低,HDL-C水平升高,HTF中剂量组大鼠TC水平降低(P<0.05).与空白组比较,模型组勃起次数、ICP/MAP、血清HDL-C、阴茎海绵体组织NO含量降低,血清TC、LDL-C、VEGF、ET-1水平升高(P<0.05).与假手术组比较,模型组勃起次数减少,血清VEGF、ET-1水平升高(P<0.05).与模型组比较,西地那非组血清ET-1水平降低,阴茎海绵体组织NO含量升高(P<0.05);HTF各剂量组大鼠血清LDL-C、VEGF、ET-1水平降低,HDL-C水平升高,HTF低剂量组阴茎海绵体组织NO含量升高(P<0.05),HTF中剂量组勃起次数增加、血清TC水平降低(P<0.05).与西地那非组比较,HTF各剂量组大鼠血清LDL-C、VEGF水平降低,HTF低、高剂量组大鼠HDL-C水平升高,HTF低剂量组ET-1水平降低(P<0.05).肠道菌群分析显示,HTF可影响肠道菌群结构.与空白组比较,模型组丁酸菌属(Butyricicoccus)、瘤胃球菌属(Ruminococcus)、普雷沃氏菌科 UCG-001(Prevotellaceae_UCG-001)、厌氧棍状菌属(Anaerotruncus)丰度升高,Candidatus_Stoquefichus属、费克蓝姆菌属(Facklamia)、耐盐咸海鲜球菌属(Jeotgalicoccus)、肠球菌属(Enterococcus)丰度降低(P<0.05).与模型组比较,HTF中剂量组葡萄球菌属(Staphylococcus)、普拉梭菌属(Faecalibacterium)、耐盐咸海鲜球菌属、嗜盐单胞菌(Halomonas)、Negativibacillus 属、无色杆菌属(Achromobacter)、Subdoligranulum属、毛螺菌科 UCG-010(Lachnospiraceae_UCG-010)、嗜胆菌属(Bilophila)、Nosocomiicoccus 属丰度升高(P<0.05),粪杆菌属(Faecalibaculum)、Family_ⅩⅢ_UCG-001 属、毛螺菌科 UCG-006(Lachnospiraceae_UCG-006)、丁酸菌属、unidentified_Ruminococcaceae 属、毛螺菌科 ND3007(Lachnospiraceae_ND3007_group)、乳球菌属(Lactococcus)丰度降低(P<0.05).结论 化湿通痹方可能通过影响肠道菌群丰度与多样性,改善血脂异常和血管内皮细胞功能,提高大鼠勃起功能.
Effects of Huashi Tongbi Formula on Blood Lipid Levels,Vascular Endothelial Cells and Intestinal Flora in Rats with Erectile Dysfunction
Objective To observe the effects of Huashi Tongbi Formula(HTF)on blood lipid levels,endothelial cell function,and gut microbiota in rats with erectile dysfunction(ED).Methods SD rats were randomly divided into blank group,sham operation group,model group,sildenafil group,and HTF low,medium,and high dose groups,with 7 rats in each group.Hyperlipidemia combined with bilateral internal iliac artery ligation were used to establish ED model.After modeling,the sildenafil group received sildenafil(4.5 mg/kg)by gavage,while the HTF group received low,medium,and high doses(3.24,6.48,12.96 g/kg)of HTF by gavage.The other three groups received an equal volume of physiological saline by gavage.After 6 weeks of intervention,the intracavernous pressure(ICP)of the penis and mean carotid artery pressure(MAP)were measured using a multi-channel physiological signal recorder,and the ICP/MAP were calculated,the number of erections in rats was recorded.Serum total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C)were measured using a fully automated biochemical analyzer.ELISA was used to detect the levels of vascular endothelial growth factor(VEGF),endothelin-1(ET-1)in serum,and microplate method was used to detect the level of nitric oxide(NO)in the corpus cavernosum tissue of rats.16 S rDNA sequencing was used to detect the gut microbiota of rats.Results Compared with baseline,the serum LDL-C levels of rats in each dose group of HTF decreased,HDL-C levels increased(P<0.05),the TC levels of rats in the medium dose group of HTF decreased(P<0.05).Compared with the blank group,the model group showed a decrease in number of erections,ICP/MAP,serum HDL-C,and NO levels in the corpus cavernosum tissue of the penis,while serum TC,LDL-C,VEGF,and ET-1 levels increased(P<0.05).Compared with the sham operation group,the model group showed a decrease in number of erections and an increase in serum VEGF and ET-1 levels(P<0.05).Compared with the model group,the sildenafil group showed a decrease in serum ET-1 levels and an increase in NO levels in the corpus cavernosum tissue of the penis(P<0.05).The levels of serum LDL-C,VEGF,and ET-1 in rats of various doses of HTF decreased,while the level of HDL-C increased(P<0.05).The low dose group of HTF had an increase in the level of NO in the corpus cavernosum tissue of the penis,while the medium dose group of HTF had an increase in the number of erections and a decrease in serum TC levels(P<0.05).Compared with the sildenafil group,the serum LDL-C and VEGF levels of rats in each dose group of HTF decreased,the HDL-C levels of rats in the low and high dose groups of HTF increased,and the ET-1 levels of rats in the low dose group of HTF decreased(P<0.05).Analysis of gut microbiota shows that HTF can affect the structure of gut microbiota.Compared with the blank group,the abundance of Butyricicoccus,Ruminococcus,Prevotellaceae_UCG-001,and Anaerotruncus increased in the model group,while the abundance of Candidatus_Stoquefichus,Facklamia,Jeotgalicoccus,and Enterococcus decreased(P<0.05).Compared with the model group,the HTF medium dose group showed an increase in the abundance of Staphylococcus,Faecalibacterium,Jeotgalicoccus,Halomonas,Negativibacillus,Achromobacter,Subdoliguranulus,Lachnospiraceae_UCG-010,Bilophila,and Nosocomicoccus(P<0.05),while Faecalibacterium,Family_ⅩⅢ_UCG-001,and Lachnospiraceae_UCG-006,Butyricicoccus,unidentified_Ruminococcaceae,Lachnospiraceae_ND3007_group,and Lactococcus decreased(P<0.05).Conclusions The HTF may improve lipid disorders and vascular endothelial cell function in rats via affecting the abundance and diversity of gut microbiota,thereby to enhance erectile function.
