目的 探讨补骨脂提取物对去卵巢骨质疏松大鼠骨代谢指标的影响.方法 将60只雌性SD大鼠随机分为对照组、模型组、补骨脂提取物组和西药组,每组15只.除对照组外,各组通过切除大鼠两侧卵巢组织进行造模,对照组大鼠仅暴露卵巢而不行切除术.造模成功后,补骨脂提取物组给予补骨脂素(88 mg/kg)灌胃,西药组给予阿仑膦酸钠(7 mg/kg)灌胃,对照组和模型组则灌胃等体积生理盐水;连续12周.采用酶联免疫吸附实验检测各组大鼠血清骨钙素(osteocalcin,OCN)、骨特异性碱性磷酸酶(bone-specific alkaline phosphatase,BAP)、Ⅰ 型胶原交联 C-末端肽(C-terminal telopeptide of typeⅠ collagen,CTX)水平;采用RT-PCR法检测大鼠股骨和胫骨骨髓中Wnt3a和β-catenin的mRNA表达;采用双能X线骨密度仪检测大鼠腰椎和股骨的骨密度(bone mineral density,BMD).结果 与对照组比较,模型组大鼠血清CTX水平显著升高(P<0.05),OCN、BAP含量明显降低(P均<0.05);与模型组比较,各用药组大鼠血清CTX水平显著降低(P均<0.05),OCN、BAP含量明显升高(P均<0.05),但各用药组之间比较,差异无统计学意义(P>0.05).与对照组比较,模型组大鼠股骨和胫骨骨髓中Wnt3a、β-catenin的mRNA表达明显降低(P均<0.05),第4腰椎及左、右股骨BMD明显降低(P均<0.05);与模型组比较,各用药组大鼠股骨和胫骨骨髓中Wnt3a、β-catenin的mRNA表达水平,第4腰椎及左、右股骨BMD均升高(P均<0.05),但各用药组之间比较,差异无统计学意义(P>0.05).结论 补骨脂提取物能提高去卵巢骨质疏松大鼠骨密度,改善骨代谢指标,其作用可能与活化Wnt/β-catenin 有关.
Effects of Psoraleae Fructus Extract on Bone Metabolism Levels in Ovariectomized Rats with Osteoporosis
Objective To explore the effects of Psoraleae Fructus extract on bone metabolism indicators in ovariectomized osteoporosis rats.Methods Sixty female SD rats were randomly divided into control group,model group,Psoraleae Fructus extract group,and western medicine group,with 15 rats in each group.Except for the con-trol group,all other groups were modeled by removing the ovarian tissue on both sides of the rats.The control group rats only exposed their ovaries and did not undergo surgery.After successful modeling,the Psoraleae Fructus extract group was given psoralen(88 mg/kg)by gavage.The western medicine group was given alendronate sodium(7 mg/kg),while the control group and model group were given an equal volume of physiological saline.Each group was continuously gavaged for 12 weeks.The serum levels of osteocalcin(OCN),bone-specific alkaline phos-phatase(BAP),and C-terminal telopeptide of type Ⅰ collagen(CTX)in each group of rats were detected by enzyme-linked immunosorbent assay.The mRNA expression of Wnt3a and β-catenin in femoral and tibial bone marrow were detected by RT-PCR method.The bone mineral density(BMD)of the lumbar spine and femur in rats was measured by dual energy X-ray absorptiometry.Results Compared with the control group,the serum level of CTX in the model group was significantly increased(P<0.05),while the contents of OCN and BAP were significantly reduced(P all<0.05).Compared with the model group,the serum levels of CTX in each medication group were significantly reduced(P all<0.05),while the levels of OCN and BAP were significantly increased(P all<0.05).However,there was no significant difference between the medication groups(P>0.05).Compared with the control group,the mRNA expression of Wnt3a and β-catenin in the model group were significantly reduced(P all<0.05),and the BMD of the 4th lumbar spine and left and right femurs were significantly reduced(P all<0.05).Compared with the model group,the mRNA expression of Wnt3a and β-catenin and the BMD of the 4th lumbar spine and left and right femurs in each medication group increased(P all<0.05),but there was no significant difference differ-ence between the medication groups(P>0.05).Conclusion Psoraleae Fructus extract could increase bone mineral density and improve bone metabolism indicators in ovariectomized osteoporosis rats,and its effect may be related to the activation of Wnt/β-catenin.
