摘要

目的 观察活血明目方对体外加压大鼠视网膜神经节细胞(retinal ganglion cells,RGCs)中凋亡相关因子Bax和Bcl-2表达的干预作用.方法 将SD大鼠随机分为4组,每组3只大鼠,采取体外加压培养SD大鼠凋亡模型,制备活血明目方含药肠吸收液,观察活血明目方对体外加压RGCs在NF-κB信号通路作用下对Bax、Bcl-2表达的影响.结果 Q-PCR和Western blot检测发现,与模型组相比,含药肠吸收液组中Bax相对表达量显著下降(P<0.05),而Bcl-2相对表达量则显著上升(P<0.01).结论 活血明目方含药肠吸收液对RGCs凋亡具有一定的抑制作用.

Abstract

Objective To observe the intervention effect of blood-boosting and eye-opening agent on the expression of Bax and Bcl-2 genes in RGCs of rats pressurized in vitro,and to explore its mechanism of action.Methods SD rats were randomly divided into 4 groups of 3 rats each,and the apoptosis model of SD rats was cultured under in vitro pres-sure,and the intestinal absorption solution containing the medication of Live Blood and Mindfulness Formula was prepared to observe the effects of Live Blood and Mindfulness Formula on the expression of Bax and Bcl-2 by RGCs under the action of the NF-κB signalling pathway under in vitro pressure.Results Q-PCR and Western blot assays revealed that compared with the model group,the relative expression of Bax in the group containing the medicated intestinal absorption solution decreased significantly(P<0.05),while the relative expression of Bcl-2 increased significantly(P<0.01).Conclusion The blood-boosting and eye-opening agent can protect the visual function of rats pressurized in vitro and has a certain in-hibitory effect on optic nerve apoptosis.
