Study on Optimization of Refining Process of Column Chromatography of Ginkgo biloba Leaves Extracts by Response Surface Methodology
Objective:In order to get a higher content of Ginkgo biloba leaves extracts,Response Surface Methodology (RSM) are employed to optimize the refining process of column chromatography of Ginkgo biloba leaves extracts.Methods:Based on the single factor experiments,Box-Behnken design was applied to establish the mathematical model.The optimum refining process was investigated by response surface methodology with low ethanol concentration,high ethanol concentration and pH value of solution as influencing factors,and the content of total flavone and lactone were considered as index.Results:The results of RSM suggested that under the experimental conditions of low ethanol concentration 18%,high ethanol concentration 67%,pH value 5.8.The average product content of total flavone and lactone were 29.50% and 10.58% under above conditions.Conclusion:This optimized technology of column chromatography of Ginkgo biloba leaves extracts by response surface methodology is stable,and feasible,which could be extended to the applications of large-scale production.
