UPLC Specific Chromatogram of Compound Luobuma Granules and Content Determination of Six Components
Objective:To establish a ultra performance liquid chromatography(UPLC)for Compound Luobuma Granules,and neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,hyperoside,isoquercitrin and cynaroside were determined.Methods:Gradient elution was used with Waters CORTECS T3(100 mm×2.1 mm,1.6 μm)column and acetonitrile-0.1%phosphoric acid as mobile phase at a flow rate of 0.33 mL·min-1.The detection wavelength was 360 nm,and column temperature was 30℃.Results:The specific chromatogram of 15 batches of Compound Luobuma Granules were established,from which 14 characteristic peaks were identified,with all the similarities more than 0.95.Neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,hyperoside,isoquercitrin and cynaroside all had good linear relationships with the peak area in their corresponding ranges(r≥0.999 9).The average recoveries of the six components were 99.9%,100.8%,99.9%,97.6%,101.1%,and 100.6%,respectively,with RSD<2.0%.Conclusion:This developed UPLC had high sensitivity and good reproducibility,which provided a basis for scientific evaluation and comprehensive control of the quality of Compound Luobuma Granules.
