Detection of Fungi in Seeds of Astragalus membranaceus var.mongholicus and Screening of Seed Coatings
Objective:To screen suitable seed coatings for Astragalus membranaceus var.mongholicus by detecting the external fungal spore count and internal fungal species.Methods:The plate culture method was employed to detect external fungal spore count and internal fungal species in 10 batches of A.membranaceus var.mongholicus seeds collected from different areas.The fungi were isolated and identified.The germination boxes and potato dextrose agar(PDA)were used to examine the antifungal effects of the combinations of four common seed coatings.Results:The count of fungal spores showed significant differences among the seeds from different areas(P<0.05),being the highest on the seeds from Longxi County in Gansu Province and the lowest on the seeds from Wuzhai County in Shanxi Province.Fungi of Penicillium and Alternaria were detected in the seeds.The combinations of seed coatings showed varied effects on the seeds from different areas.Specifically,the seed coating combination of Manyijia+Ruisheng exhibited strong antifungal effect on the seeds from Longxi County in Gansu Province but poor effects on the seeds from Minle County in Qinghai Province and Wuzhai County in Shanxi Province.Linong/Dingmiaoxin+Ruisheng demonstrated strong antifungal effects on the seeds from Yingxian County in Shanxi Province,Minle County in Gansu Province,and Huzhu County in Qinghai Province but limited effects on the seeds from other areas.Conclusion:This study highlights the need for formulating specific seed coating combinations tailored to the seeds of A.membranaceus var.mongholicu in different producing areas.
