Interference Factors and Countermeasures in MTT Assay for Detection of Chinese Herbal Medicine Components with Cytotoxicity and Influencing Cell Proliferation
The methyl thiazolyl tetrazolium(MTT)assay with simple operation and low cost has been widely used in the research on the Chinese herbal medicine components which have cytotoxicity and influence cell proliferation.However,the test conditions such as the number and density of cells,MTT concentration,and reaction duration interfere with the accuracy of the test results.In addition,the diverse components with complex structures in Chinese herbal medicines may affect the accuracy of the detection results due to the drift of the absorption spectrum,interference of cellular metabolic activity,absorption peaks of approximate detection wavelengths,which result in erroneous conclusions.By reviewing the relevant articles published in recent years,the paper summarizes and analyzes the interference factors in test conditions and corresponding optimization measures and quality control measures.Furthermore,this paper classifies some Chinese herbal medicine components that need attention in the assay,summarizes the alternative methods of MTT assay and the application of MTT assay combined with other assays,and discusses the existing problems in the application,aiming to provide reference for the rational application of MTT assay in the research on Chinese herbal medicine components.
