Analysis of Chemical Components in Stem and Tuber of Gastrodia elata Bl.f.elata"Luema No.1"by UPLC-Q-TOF-MS/MS
Objective:To determine and compare the chemical components in the stem and tuber of Gastrodia elata Bl.f.elata"Luema No.1".Methods:Ultra-performance liquid chromatography-quadrupole/time-of-flight tandem mass spectrometry(UPLC-Q-TOF-MS/MS)was performed with the Xbridge BEH C18 column(100 mm×2.1 mm,1.7 μm)and the mobile phases of 0.1%formic acid aqueous solution-acetonitrile.The gradient elution was conducted at a flow rate of 0.3 mL·min-1,and the data were collected by Full-ddMS2 in positive and negative ion modes. The reference substances, related literature, and mass spectrometry(MS)fragmentation patterns were taken as reference to analyze the MS/MS data of the components in the samples. Results:A total of 153 compounds were identified in the stem of"Luema No.1",including 20 phenols,46 organic acids,11 flavonoids,12 amino acids,14 alkaloids, and 50 other compounds.A total of 126 compounds were identified in the tuber of"Luema No.1",including 21 phenols,36 organic acids,13 amino acids,11 alkaloids, and 45 other compounds. Conclusion:The stem and tuber of"Luema No.1"showed different material bases and should be distinguished in the development and application. The stem demonstrates higher development and application value.
