Quality Evaluation of Akebiae Frucus[Akebia trifoliate(Thunb.)Koidz.]Dispensing Granules by Combination of Fingerprint,Multi-Component Quantitative Analysis,and Chemical Pattern Recognition
Objective:To establish a quality evaluation method for Akebiae Frucus[A.trifoliate(Thunb.)Koidz.]dispensing granules by combining fingerprinting,multi-component quantitative analysis,and chemical pattern recognition.Methods:Standard Akebiae Frucus[A.trifoliate(Thunb.)Koidz.]decoctions and dispensing granules were prepared.High-performance liquid chromatography(HPLC)fingerprinting was established,and the contents of eight components were determined.The transfer rate and extraction rate were calculated,and similarity evaluation,cluster analysis(HCA),principal component analysis(PCA),and partial least squares discriminant(PLS-DA)were performed to evaluate the consistency of chemical components between dispensing granules and standard decoctions.Results:The mass fractions of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,calceolarioside B,isochlorogenic acid B,isochlorogenic acid A,isochlorogenic acid C,and akebia saponin D in standard Akebiae Frucus[A.trifoliate(Thunb.)Koidz.]decoctions were 1.94-10.33,2.18-13.77,1.77-9.15,2.31-19.34,1.77-6.48,1.16-5.55,2.55-9.90,and 0.92-8.93 mg·g-1,respectively.The transfer rates of these components were 36.25%-56.02%,24.64%-40.23%,35.88%-58.41%,29.45%-50.69%,16.55%-29.54%,26.25%-46.59%,25.33%-41.66%,and 11.03%-17.35%.The extraction rate was 13.59%-24.49%.The component content,transfer rate,and extraction rate of Akebiae Frucus[A.trifoliate(Thunb.)Koidz.]dispensing granules were close to those of standard decoctions.The results of similarity evaluation,HCA,PCA,and PLS-DA indicated good consistency of chemical components between dispensing granules and standard decoctions.Conclusion:There was good consistency between dispensing granules and standard decoctions of Akebiae Frucus[A.trifoliate(Thunb.)Koidz.].The results were reasonable and reliable,which can provide a reference for process optimization and quality evaluation.
