Quality Evaluation of Ajuga decumbens from Different Producing Areas Based on UPLC Characteristic Chromatogram and Content Determination
Objective:To establish the characteristic chromatogram of Ajuga decumbens Thunb.by ultra-high performance liquid chromatography(UPLC)and method for determining the content of harpagoside and acetyl harpagoside.Methods:Waters BEH C18(150 mm×2.1 mm,1.7 μm)column was used.The mobile phase A was acetonitrile-methanol(4:1),and the mobile phase B was 0.2%phosphoric acid aqueous solution for gradient elution.The flow rate was 0.30 mL·min-1,and the column temperature was 30℃.The detection wavelength was 202 nm,and the injection volume was 2 μL.The characteristic chromatogram of A.decumbens was established to determine the content of harpagoside and acetyl harpagoside.Results:A total of 10 common fingerprint peaks were identified in the characteristic chromatograms of 17 batches of A.decumbens,among which four components were identified as harpagoside,acetyl harpagoside,luteonin-7-diglucosidic acid,and apigenin-7-diglucosidic acid.The mass fraction of harpagoside in the 17 batches of A.decumbens.was in the range of 0.33%-0.95%,while that of acetyl harpagoside was 0.47%-1.93%.Conclusion:The established UPLC characteristic chromatogram and the method for determining the content of harpagoside and acetyl harpagoside can provide a basis for the quality evaluation of A.decumbens.
